| Zhenwu Decoction is a compound preparation consisting of aconite,peony,atractylodes,alum,and ginger in a ratio of 3:3:2:3:3.The clinical application is quite extensive.Because of its strong cardiac effect,it is important for treating heart failure.One of the prescriptions.At present,the research on Zhenwu Decoction mainly focuses on pharmacological activity and clinical application.The research on the material basis is still insufficient.Therefore,studying the extraction process of Zhenwu Decoction and testing its chemical composition can help to clarify its active ingredients and clarify its material basis to lay the foundation for more in-depth research.OBJECTIVE:In view of its extensive clinical role in the treatment of heart failure,this study is based on the optimal extraction process of Zhenwu Decoction based on the index component of Zhenwu Decoction,in order to improve its clinical utility.By establishing the chemical composition information base of aconite,medlar,atractylodes,peony and ginger in the composition of Zhenwu Decoction,using UPLC/Q-TOF-MS,UPLC/Q-TOF-MS The compound was tested and the UNIFI analysis platform was used to comprehensively analyze the chemical composition of Zhenwu Decoction.Methods:(1)The subject was investigated by dry paste rate,aconitine,paeoniflorin,atractylenolide III and norepinephrine as indicators,because aconitine was a toxic component in the indicator component.The low is better,and the dry paste rate and other effective indicators are high,so this experiment uses a comprehensive score:that is,the subtraction method is used to score the aconitine based on the traditional score,and the single factor test is used to investigate.The optimal extraction process parameters are optimized in combination with the orthogonal experimental design.(2)This project used UPLC/Q-TOF-MS to test the samples of Zhenwu Decoction and its prescriptions,and qualitatively analyzed the results using UNIFI software.First,select C18 reversed-phase column,ACQUITY UPLC CSH C18(100 mm×2.1 mm,1.7μm),with acetonitrile-0.1%formic acid as mobile phase,gradient elution,detection wavelength 230 nm,column temperature 40°C,flow rate 0.2 mL/min,injection volume 10μL,the chromatographic method of UPLC chromatogram of Zhenwu Decoction was established;secondly,it was combined with quadrupole time-of-flight mass spectrometry(Q-TOF-MS)in positive and negative ion modes.Detecting and obtaining the exact molecular weight and molecular formula of each component;finally,analyzing the mass spectrum by UNIFI software,analyzing the cleavage information of each component,and analyzing and identifying the chemical components of Zhenwu Decoction in combination with the literature.Results:(1)The subject identified the best extraction process factor combination A3B2C3,namely:adding 10 times the amount of water,boiling 2 times,extraction time 2h.(2)This study identified 71 possible components in Zhenwu Decoction by negative ion mode,and elaborated the primary and secondary mass spectrometry and mass spectrometry fragmentation pathways of 21 of them,2 of which were derived from aconite and 4 were derived from poria cocos,2 are from Atractylodes,5 are from white peony,and 8 are from ginger.(3)By comprehensively analyzing the mass spectrograms of Zhenwu Decoction,Zhenwu Decoction serum and blank serum,21prototype components in the plasma of Zhenwu Decoction were identified.Conclusions:The subject successfully optimizes the optimal extraction process parameters,and uses UPLC/Q-TOF-MS combined technology to carry out the analysis and identification of the chemical constituents of Zhenwu Decoction,and comprehensively clarifies the chemical composition of Zhenwu Decoction,in order to find out the pharmacody-namic material basis of Zhenwu Decoction,and to Zhenwu The further research of the soup provides a scientific basis. |
