| [Research background and Objective]Liuwei dihuang pill(LW) is a classic traditional chinese medicine(TCM)formula consisting of six herbs: Radix Rehmanniae(Dihuang;prepared root of Rehmannia glutinosa),Rhizoma dioscoreae(Shanyao;rhizome of Dioscorea opposita),Fructus corni(Shanzhuyu;fruit of Cornus officinalis),Cortexmoutanradicis(Mudanpi;root bark of Paeonia suffruticosa),Rhizoma Alismatis(Zexie;rhizome of Alisma plantago-aquatica),and Poria(Fuling;sclerotia of Poria cocos).The results of our studies have shown that LW exerts a broad spectrum of pharmacological effects,such as ameliorating the decline in learning and memory and the decline in function related to aging processes and geriatric diseases,as well as enhancing the estrogenic activity during menopause.LW also exerts anti-osteoporotic,antihypertensive,anti-inflammatory,antitumor,and antioxidant effects.In this study,we established an inflammatory model of the central nervous system,to explore whether LW can improve the learning and memory impairment induced by center imflammation through regulating center glycometabolism in rats.[Methods]Novel object location test,novel object recognition test and Y maze test were used to measure the learning and memory induced by chronic variate stress co-treated with LPS.The effect of chronic variate stress co-treated with LPS to IL-1β and TNF-α was measured by western blot(WB).The effect of LW on chronic stress co-treated with LPS-induced increased was measured by WB.The effect of chronic variate stress co-treated with LPS to hippocampal insulin signaling pathway was measured by WB.The effect of LW on chronic stress co-treated with LPS-induced abnormal of hippocampal insulin signaling pathway was measured by WB.[Results]The decrease of discrimination index of novel object location test and novel recognition test,correct alternation rate of Y maze induced by chronic variate stress co-treated with LPS was ameliorated by LW.The increased of IL-1β and TNF-α in hippocampus of rats induced by chronic variate stress co-treated with LPS was ameliorated by LW.The expression of p IRS1 was down-regulated and cell menbrane Glu T1 were up-regulated of rat hippocampal induced by chronic stress co-treated with LPS was ameliorated by LW.[Conclusion]LW ameliorates chronic stress co-treated with LPS-induced learning and memory impairment through mediating center glycometabolism in rats. |
