| BackgroundPrimary immune thrombocytopenia(ITP)is a disease characterized by thrombocytopenia and characterized by skin or mucosal bleeding.TCM further subdivides ITP into four syndromes:blood-heat delusion,Qi failing to control blood,blood stasis obstructing collaterals,Yin deficiency and fire prosperity.The treatment of ITP is often based on the specific conditions of patients.At present,ITP-related drug research mainly relies on single ITP models,such as ITP model established by injecting monoclonal/polyclonal antibodies or platelets.ITP model is simple and effective,but it has many limitations.Compared with it,ITP disease-syndrome combination model is more suitable for complex traditional Chinese medicine research.In addition,most of the studies are about the chemical composition,clinical efficacy,mechanism of action,etc.There are relatively few theoretical studies on Qi and blood by disassembling prescriptions.This paper is based on the model of ITP QBSX,to study the YQSX Granule and its decomposition,and try to explore the relationship between Qi and blood.1 Objective1.1 Effects of YQSX Granule,Qi-tonifying Drug and Blood-nourishing Drug on ITP QBSX Model.1.2 Initially explore the relationship between Qi and blood in QBSX syndrome.2 Method2.1 CBA/J mice were randomly divided into normal group,model group,low-dose group,high-dose group and prednisone acetate(PA)group.Except normal group,the rest four groups were deprived of sleep at the first week,deprived of sleep at the second,third and fourth weeks and injected with rat platelets intraperitoneally.From the first day of modeling,the drug groups were given low dose YQSX Granule(15.0g raw drug/kg),high dose YQSX Granule(30.Og raw drug/kg)and prednisone acetate(0.75mg/kg),respectively.The platelet count was measured weekly and the mice were weighed at 0,1,4 and 5 weeks respectively.2.2 CBA/J mice were randomly divided into normal group,model group,QBSX group,Qi-invigorating group,blood-nourishing group and prednisone acetate(PA)group.Except the normal group,the rest five groups were deprived of sleep in the first week,deprived of sleep in the second,third and fourth weeks,and injected with rat platelets intraperitoneally.From the first day of modeling,the drug group was given YQSX Granules(15.Og crude drug/kg),Qi-tonifying drugs(5.7g crude drug/kg),blood-nourishing drugs(9.3g crude drug/kg)and prednisone acetate(0.75mg/kg),respectively.Body mass,grip,spontaneous,tongue,platelet count,MPV,platelet apoptosis,red blood cell count,hemoglobin,T cell subsets,thymus and spleen index were measured.2.3 Balb/c mice were randomly divided into normal group,model group,QBSX group,Qi-invigorating group and blood-nourishing group.Except for the normal group,the other four groups were injected APS every other day except sleep deprivation in the first week and in the second and third weeks.The administration mode was the same as 2.2.Body mass,grip,pulse,platelet count,MPV,red blood cell count,hemoglobin determination,T cell subsets,Breg cells and spleen index were measured.HE staining pathological sections of liver,spleen and abdominal aorta vessels were observed.3 Research result3.1 The body weight and platelet count of the model group were significantly lower than those of the normal group(P<0.01).There was no significant difference in body mass between the drug group and the model group,and the platelet count increased significantly compared with the model group(P<0.05).3.2 In the model group,the skin was rough and dull,the spirit was depressed,the tongue was pale,the body weight,grip,exercise distance,platelet count,red blood cell count,hemoglobin determination,CD3+,CD3+CD4+,CD3+CD8+,Treg cells were significantly lower than those in the normal group.Grasp force,platelet count,erythrocyte count,hemoglobin determination,CD3+,CD3+CD4+,CD3+CD8+,Treg cells in YQSX group were significantly higher than those in model group.Grasp,platelet count,erythrocyte count and hemoglobin in the Qi-supplementing group were significantly higher than those in the model group.The platelet count and red blood cell count in the blood-nourishing group were significantly higher than those in the model group.3.3 In model group,the skin of mice was rough and dull,and their spirit was depressed.The body weight,grip,pulse amplitude,platelet count,red blood cell count,hemoglobin determination,CD3+,CD3+CD4+,CD3+CD8+were significantly lower than those in normal group,while the Breg cell and spleen index were significantly higher.Grasp,pulse amplitude and platelet count were significantly increased and Breg cells were decreased in the YQSX group.Grasp,pulse amplitude,erythrocyte count,CD3+,CD3+CD4+,CD3+CD8+increased significantly and Breg cells decreased in the Qi-supplementing group.Grasp,pulse amplitude,platelet count,erythrocyte count,hemoglobin measurement,CD3+,CD3+CD4+,CD3+CD8+increased significantly in the blood-nourishing group,while Breg cells decreased.4 Conclusion4.1 YQSX Granule and its disassembled prescriptions can improve both the Qi deficiency syndrome and the blood deficiency syndrome of ITP.4.2 The results showed that the effects of the two formulations were similar,but there were differences,which needed further study. |
