| Objective: To study the level of autophagy and the expression of PI3K-AKT-m TOR signaling pathway in asthmatic rats with syndrome of cold retention accumulation in lung,and to explore the role of autophagy and PI3K-AKT-m TOR signaling pathway in the pathogenesis of the syndrome.By observing the relative indexes,we explore the pathological manifestations of “lung yang deficiency” in the pathogenesis of asthma with syndrome of cold retention accumulation in lung and further clarify the therapeutic mechanism of Wenyanghuayin Decoction(Xiaoqinglong Decoction plus ginseng and astragalus)on asthmatic cold drink and lung syndrome.We hope to provide a basis for scientifically and rationally establishing rules and guiding clinical medication.Methods: Methods of theoretical research and experimental research are adopted.With the theory of qi,yin and yang,etiology and pathogenesis,we elaborated on “lung yang” and “lung yang deficiency”,aiming to enrich Chinese medicine theory.In the experimental research,we adopted HE staining pathological sections,PCR fluorescence detection,Western Blot and other methods.We also determined the pathological mechanism and related inflammatory factors of PI3K-AKT-m TOR signaling pathway associated with the treatment of asthma with syndrome of cold retention accumulation in lung,and improved the research of astragalus polysaccharide,ginseng saponin,Xiaoqinglong decoction and Wenyanghuayin formula for the treatment of sthma with syndrome of cold retention accumulation in lung,and probed into the new model of asthmatic rats with syndrome of cold retention accumulation in lung.And with the micro-indicators,we compared the treatment mechanism of Wenyang Huayin Decoction,Ginsenoside,Astragalus Polysaccharide and Xiaoqinglong Decoction for asthma with syndrome of cold retention accumulation in lung.Results: Through theoretical analysis of lung yang and lung yang deficiency,we confirmed the existence of “lung yang”.It was found that “lung yang deficiency” will further affect the level of autophagy,which will reduce the level of autophagy and increase the level of airway inflammation,leading to the occurrence or worsening of asthma with syndrome of cold retention accumulation in lung.Furthermore,it was confirmed that Wenyang Huayin formula can act on the PI3K-AKT-m TOR signaling pathway,and regulate the autophagy-related proteins such as ATG and LC3 to make the autophagy level normal.This may be the main mechanism of Wenyanghuayin formula in treating asthma.From the microscopic point of view,it was found that the HE staining pathological section,RT-PCR fluorescence quantitative detection,Western blotting detection and bronchial-related inflammatory factors and other related indexes of the rats in the asthma group were significant.Compared with the model group,the Astragalus polysaccharide group,ginseng saponin group,Xiaoqinglong decoction group and Wenyanghuayin decoction group have obvious differences in morphology and physical signs,and various microscopic indicators also have also improved in different degrees.Conclusion: From the theoretical point of view,the physiological functions of “lung yang” can be summarized into the aspects of warming the body,defending the external pathogens,spreading and descending lung qi,as well as regulating water metabolism.Lung yang is closely related to yang qi of other organs physiologically and pathologically.Through experimental studies,it was confirmed that Wenyang Huayin Decoction can affect the autophagy-related genes of ATG and LC3 by regulating the PI3K-AKT-m TOR signaling pathway,and significantly improve the autophagy level of lung tissue in rats with syndrome of cold retention accumulation in lung.It has a certain therapeutic role in treating asthmatic cold.In addition,the expression of autophagy gene and the related protein expression of PI3K-AKT-m TOR signaling pathway in rat lung tissue further confirmed that Wenyanghua Decoction is superior to Xiaoqinglong Decoction in treating asthma with syndrome of cold retention accumulation in lung.And the reason is that polysaccharide and ginseng saponin regulate the autophagy and inflammation of lung tissue in asthmatic cold drink syndrome,which also proves that the combination of multi-flavor drugs can exert the best therapeutic effect.It has great significance for the development of Chinese medicine theory and the clinical treatment of asthma with syndrome of cold retention accumulation in lung. |
