| ObjectiveThe medical records of professor Yu shaoyuan in the treatment of chronic atrophic gastritis in the past three years were selected,The Excel was used for the statistics of traditional Chinese medicine(TCM)syndromes and frequency of TCM,and the characteristics of professor Yu Shaoyuan’s prescription for the treatment of CAG with the method of strengthening spleen,removing blood stasis and detoxifying were summarized.The research object is the internationally recognized rat model of CAG,a variety of pharmacodynamic indexes were used to observe the therapeutic effect of WeipiXiao on spleen-deficiency type of CAG rats,and to explore the mechanism of action,so as to provide data support for the next clinical drug development and provide a new idea for the treatment of CAG with TCM.Methods1.Study on characteristics of formulaeThe medical cases diagnosed with CAG by professor Yu Shaoyuan from January 2016 to December 2018 were selected,and all the collected clinical data were sorted out and screened.By using Excel to make statistics of TCM syndromes and TCM frequency,the clinical characteristics of professor Yu Shaoyuan in the treatment of CAG were summarized2.Research on the effect mechanismSixty SD rats(SPF grade,The males and females were evenly split)were divided into blank control group and model group by random number table method.Model group were fed with 160 μg/mL of MNNG water,irregular diet,Lavage 40%ethanol and Ranitidine’ s solution to establish model rats of CAG for 8 consecutive weeks.After 8 weeks,3 rats were randomly selected and sacrificed to death under anesthesia.The atrophy of the entire layer of 1.5cm X 1.5cm gastric antrum was observed by microscopy,and the model was judged to be successful,and than model group were divided into model group,Weipixiao group and Weifuchun group.At the beginning of 9th week,the rats of Weipixiao group and Weifuchun group were provided with treatment for 4 weeks,and observe curative effects of Weipixiao on gastric mucosa in rats from the point of gastric and changes of pathological and histologicalResult1.Study on characteristics of formulaeThrough the analysis of professor Yu Shaoyuan’ s 74 patients with CAG and 189 outpatient prescriptions from 2016 to 2018.A total of 79 drugs were included.Chenpi,muxiang,dangshen,sharen,taizishen,huangqi,wuzhimaotao,ba ihuasheshecao,banzhilian,maiya,daoya,zisugen,fuling,sanqi,haipiaoxiao,zh ebeimu,pugongying,zhike,zhuru,houpo,yujin and other commonly used drugs,mainly including strengthening spleen and qi,clearing heat and dampness,and activating blood and removing blood stasis.2.Research on the effect mechanism(1)Effect on gastric secretion in rats with CAG Compared with the normal group,the pH value of gastric juice in the model group was significantly increased(P<0.01),and the pepsin activity and free gastric mucus content were significantly reduced(P<0.01).The pH of gastric juice in Weipixiao group increased(P<0.05).The pH value of gastric juice in Weifuchun group was significantly increased(P<0.01).Compared with the model group,the ph of gastric juice in the weipixiao group was significantly decreased(P<0.01),and the pepsin activity was increased(P<0.05).Gastric juice pH value decreased in weifuchun group(P<0.01),pepsin activity increased(P<0.05),free gastric mucus content increased significantly in Weipuxiao group and Weifuchun group(P<0.01),and pepsin activity increased in Weipuxiao group compared with Weifuchun group,but there was no statistical difference.(2)Effect on serum gastrointestinal hormone content in CAG rats Compared with the normal group,the serum VIp and TGF-a level in the model group was increased(P<0.05),the content of pGI、GH、pGE2 in serum decreased(P<0.01).Compared with the model group,the serum VIp and TGF-a in the Weipixiao group and Weifuchun group were decreased(P<0.05),the content of pGI、GH、pGE2 in serum increased we found no obvious difference in the normal group,model group,Weipixiao group and Weifuchun group about the content of serum PGⅡ(P>0.05).(3)Effects on gastric mucosa of rats with CAG Compared with the normal group,the gastric mucosa in the model group was significantly thinner(P<0.01),the mucosa muscle layer was significantly thicker(P<0.01),and the number of glands was significantly reduced(P<0.01).Compared with the Weifuchun group,the mucosal layer was thicker,the mucosal muscular layer was thinner,and the glands were increased in the Weipixiao group,but there was no statistical difference(P>0.05).Conclusion1.Study on characteristics of formulaeIn the treatment of CAG with TCM,professor Yu Shaoyuan takesstrengthening spleen,removing blood stasis and detoxifying" as the major method,supplemented by methods of clearing heat,supplementing deficiency,regulating qi and dampness.Dispelling pathogenic factors and balancing vital qi,constantly caring for the spleen and stomach in the middle jiao,and paying attention to the regulation of qi.2.Research on the effect of mechanismAfter 4 weeks of treatment,Weipi xiao had a clear effect on SD rats with chronic atrophic gastritis,and there was no statistically significant difference between Weipi xiao and Weifuchun.Weipixiao can promote the secretion of gastric juice,increase the acidity of gastric juice,reduce the contents of serum pGI and PGE2,reduce the contents of serum VIP and TGF-a,increase the activity of pepsin,reduce the atrophy of glands and increase the weight of rats. |
