Objective:To observe the effect of heat-sensitive moxibustion "Zhongwan"(CV12)on the expression of Wnt2,?-catenin and matrix metalloproteinase-7 in gastric mucosa of rats with chronic atrophic gastritis.The possible mechanism of heat-sensitive moxibustion in the treatment of CAG was discussed in order to provide experimental basis for the treatment of CAG with heat-sensitive moxibustion.Method:66 male SD rats were randomly divided into blank group and model group.The model group was given 100ug/mL MNNG free drinking,30%ethanol and 2% sodium salicylate mixed solution was given intragastrically and combined with hunger-satiety method to establish CAG model for 12 weeks.After the establishment of the model,the model group was randomly divided into three groups: model group,vitacoenzyme group,moxibustion group.Moxibustion group moxibustion at“Zhongwan”,once a day,40 minutes per time.The tail temperature of rats was measured and recorded per 5min.According to the changes of tail temperature of rats,the rats were divided into non-heat sensitive moxibustion group and heat-sensitive moxibustion group.The vitacoenzyme group was perfused with vitacoenzyme according to 2ml/kg/d,once a day,28 d.The pathological changes of gastric mucosa were observed by light microscope,the expression of Wnt2 protein was detected by Western Blot and immunohistochemical method,and the expression of ?-catenin,MMP-7protein was detected by immunohistochemical method.Result:1.Before the establishment of the model,all the rats in eachgroup were in good spirits and generally in good condition.After modeling,there was no significant change in blank group and before modeling.Each model group appears mental dispirited,binocular achy,body emaciated,auricle pale,lack of blood color.Slow reaction,loose stool,etc.Before modeling,the body weight of each group was at the same baseline level.After modeling,the body weight of the blank group was normal,and the weight of the rats in the model group was lower than that of the blank group(P<0.01).After the establishment of the model,no obvious abnormal gastric mucosa was observed in the blank group.In the model group,dark red or gray could be seen in the naked eye of the gastric mucosa in the model group,and the mucosal layer became thinner,elasticity decreased,fold flattened,mucus decreased,and the furrow gyrus became shallow or disappeared.The muscle layer became thinner;Under light microscope,the gastric mucosa layer became thinner,hyperemia was obvious,glands became smaller,atrophy and even disappeared,disordered arrangement,decrease in number,cystic dilatation,dysplasia and so on.2.Twenty-eight days after treatment,all the treatment groups showed different levels of mental improvement,among which the heat-sensitive moxibustion group was the most obvious improvement.After treatment,the body weight of each group increased significantly(P<0.01),and after treatment,compared with the blank group,the body mass of the model group increased slowly(P<0.01).Compared with the model group,the body mass of the non heat-sensitive moxibustion group and the vitacoenzyme group increased significantly(allP < 0.05),but there was no significant difference between the non heat-sensitive moxibustion group,the heat-sensitive moxibustion group and the vitacoenzyme group.There were no obvious pathological changes in the gastric mucosa in the blank group under light microscope,and in the model group,no obvious pathological changes were observed in the gastric mucosa.Glands of different sizes,disorder,gland atrophy,upper mucosa occasionally can be seen intestinalmetaplasia.The surface of gastric mucosa in the treatment group was better than that in the model group.The gastric mucosa glands in the heat-sensitive moxibustion group were arranged regularly,and there was no obvious abnormality.3.Compared with the blank group,Wnt2 protein expression increased in model group,non heat-sensitive moxibustion group,heat-sensitive moxibustion group and vitacoenzyme group(all P<0.01).Compared with the model group,Wnt2 protein expression was decreased in the non heat-sensitive moxibustion group,heat-sensitive moxibustion group and vitacoenzyme group(P < 0.01).Compared with the non heat-sensitive moxibustion group,the expression of Wnt2 protein in the heat-sensitive moxibustion group and the vitacoenzyme group decreased(allP <0.05).There was no significant difference between heat-sensitive moxibustion group and vitacoenzyme group.Compared with the blank group,the density mean of Wnt2 increased in the model group and the non heat-sensitive moxibustion group(P<0.01),and there was no significant difference between the heat-sensitive moxibustion group,the vitacoenzyme group and the blank group.Compared with the model group,the density mean of Wnt2 in the heat-sensitive moxibustion group and the vitacoenzyme group decreased(allP<0.01),and there was no significant difference between the non heat-sensitive moxibustion group and the model group.Compared with the non heat-sensitive moxibustion group,the density mean of Wnt2 decreased in the heat-sensitive moxibustion group and the vitacoenzyme group(P<0.05).There was no significant difference between heat-sensitive moxibustion group and vitacoenzyme group.4.Compared with the blank group,the density mean of ?-catenin in model group,non heat-sensitive moxibustion group,heat-sensitive moxibustion group and vitacoenzyme group increased(P < 0.01,P <0.05).Compared with the model group,the adensity mean of ?-catenin in the non heat-sensitive moxibustion group,the heat-sensitive moxibustiongroup and the vitacoenzyme group decreased(all P<0.01).Compared with the non heat-sensitive moxibustion group,the density mean of ?-catenin in the heat-sensitive moxibustion group and the vitacoenzyme group decreased(P<0.05,P<0.01).There was no significant difference between heat-sensitive moxibustion group and vitacoenzyme group.Compared with the blank group,the density mean of MMP-7 increased in the model group,non heat-sensitive moxibustion group,heat-sensitive moxibustion group and vitacoenzyme group(allP<0.01).Compared with the model group,the density mean of MMP-7 in the heat-sensitive moxibustion group and the vitacoenzyme group decreased(P<0.01),and there was no significant difference between the non heat-sensitive moxibustion group and the model group.Compared with the non heat-sensitive moxibustion group,the MMP-7density mean of the heat-sensitive moxibustion group and the vitacoenzyme group decreased(P<0.05).There was no significant difference between heat-sensitive moxibustion group and vitacoenzyme group.Conclusion:(1)Heat-sensitive moxibustion can significantly improve the gene ral state(such as body weight,mental state,stool character)and pa thological changes of gastric mucosa in CAG rats.The results show th at heat-sensitive moxibustion is effective in treating CAG by morphol ogical and pathological changes.(2)Heat-sensitive moxibustion can inhibit Wnt/?-catenin signaling pathway,decrease the expression of Wnt2,?-catenin,MMP-7 protein in gastric mucosa of CAG rats,and regulate invasion,invasion and metastasis of gastric mucosal cells,so as to achieve the purpose of treating CAG.
