Object:To observe the changes of??T cells and related lymphocytes in patients with chronic brucellosis before and after antimicrobial treatment,and understand the expression of these cells in patients with chronic brucellosis.Further analysis the effect of these cells on the immune status of in patients with chronic brucellosis,and to provide a theoretical basis for targeted immunotherapy of brucellosis.Methods:A total of 88 patients with chronic brucellosis were collected from the Infectious Disease Hospital of Tianjin from September 2012 to April 2018,with 68 males and20 females.The diagnostic criteria,staging criteria and antimicrobial treatment plan of all patients were based on the relevant provisions of the 2012 edition guidelines for brucellosis diagnosis and treatment.Antimicrobial treatment lasted for 6 weeks as a course of treatment.The absolute counts of??T cells,CD3~+T cells,CD4~+T cells and CD8~+T cells in peripheral blood were measured by flow cytometry.And thirty healthy volunteers were selected as healthy control group.Analysis changes of immune status of??T cells and related lymphocytes in patients with chronic brucellosis before and after treatment.And compare the immune status of??T cells and related lymphocytes in patients with chronic brucellosis and healthy controls.Results:1.The patients with chronic brucellosis completed one course of antimicrobial therapy,the average numbers of??T cells,CD3~+T cells,CD4~+T cells and CD8~+T cells before treatment were 77.9/?l,1565.2/?l,766.0/?l and 714.2/?l,and the numbers after treatment were 83.2/?l,1367.2/?l,666.4/?l and 609.6/?l.The patients with chronic brucellosis completed two course of antimicrobial therapy,the numbers of above these cells before treatment were 105.1/?l,1601.5/?l,758.0/?l and 728.7/?l,and the numbers after treatment were 79.1/?l,1289.3/?l,658.2/?l and 546.5/?l.Comparisons of cell levels before and after one course of treatment,CD4~+T cells,CD3~+T cells and CD8~+T cells counts of after treatment were lower than before treatment(P<0.05).Comparisons of cell levels before and after two course of treatment,the counts of??T cells and CD8~+T cells of after treatment were lower than before treatment(P<0.01),CD3~+T cells counts of after treatment were lower than before treatment(P<0.05).2.The average numbers of??T cells,CD3~+T cells,CD4~+T cells and CD8~+T cells before treatment in chronic brucellosis were 87.6/?l,1586.8/?l,769.9/?l and 721.1/?l,while these numbers in healthy control group were 49.4/?l,1408.7/?l,761.0/?l and 595.6/?l.Comparisons of cell levels patients with chronic brucellosis before treatment and healthy control group,the levels of??T cells and CD8~+T cells in patients with brucellosis before treatment were significantly higher than those in the healthy control group(P<0.01),and the levels of CD3~+T cells were higher than healthy controls(P<0.05).Comparisons of cell levels after one course of treatment and healthy control group,the levels of??T cells after treatment were significantly higher than those in the healthy control group(P<0.01),and the levels of CD4~+T cells were lower than healthy controls(P=0.033);comparisons of cell levels after two course of treatment and healthy control group,the levels of??T cells after treatment were significantly higher than those in the healthy control group(P<0.01).Conclusion:1.The??T cells,CD3~+T cells and CD8~+T cells of chronic brucellosis patients before antimicrobial therapy were immune activation.2.After antimicrobial treatment,the levels of??T cells,CD4~+T cells,CD8~+T cells and CD3~+T cells in patients with chronic brucellosis were significantly lower than those before treatment.In the process of eliminating Brucella,the above cells showing a downward trend,which shows that the cells play an important role in the immune response against Brucella infection.Detection of the expression level of the above cells is helpful to understand the immune status of patients with chronic brucellosis and the role of the above cells in the immune response against brucellosis infection,and to provide a theoretical basis for targeted immunotherapy of brucellosis and reduce the chronic Brucellosis.
