Objective:Through the whole genome methylation sequencing analysis of nucleated cells in peripheral blood of artemisia pollen patients with allergic rhinitis,DNA methylation difference sites related to allergic rhinitis were screened to provide a basis for the study of the occurrence and development mechanism of allergic rhinitis,and provide ideas for the forensic identification of allergic sudden death.Methods:The experiment was divided into two groups: the artemisia pollen allergic rhinitis group(n =9)and the healthy control group(n = 9).Peripheral venous blood(10ml)was collected.There were3 patients with allergic rhinitis and 3 healthy controls,respectively,for whole-genome methylation sequencing analysis,including sequencing depth and coverage,comparison to genome,CG methylation statistics,differential methylation region(DMR)analysis,and differential methylation region annotation.The differentially methylated loci obtained by sequencing analysis were analyzed by random forest analysis using R language,and further significant difference loci were screened out.Six cases of artemisia pollen allergic rhinitis patients and six cases of healthy control group were screened for DNA methylation PCR verification.Results:(1)the average sequencing coverage of each sample was 91.71%,and the average sequencing depth was 11.41;the sequencing samples were compared to the genome,with an average ratio of about 78.9%.(2)through biological information analysis and R language random forest packet statistical analysis,62 DNA methylation difference sites related to artemisia pollen allergy were screenedout.(3)further limited screening was conducted to obtain 3 different DNA methylation sites related to artemisia pollen allergy..Conclusion :(1)differences exist in DNA methylation of nucleated cells in peripheral blood between artemisia pollen allergic rhinitis patients and healthy people.(2)abnormal methylation of SEC63,RUNX1T1 and SMARCA2 is involved in the pathogenesis of allergic rhinitis.
