Objective: To investigate the expression of long non-coding RNA PRSS3P2 and its related mRNAs PRSS1,PRSS2 and PRSS3 in thyroid papillary carcinoma,and to analyze their relationship with clinical features.To investigate the invasion and migration of BCPAP cells by long non-coding RNA PRSS3P2.To prove that the high expression of lncRNA-PRSS3P2 affects the invasion and metastasis of papillary thyroid carcinoma,which increases the risk of surgery and causes postoperative recurrent laryngeal nerve injury,which leads to medical disputes.Methods:(1)100 cases of fresh thyroid papillary carcinoma tissue and normal tissue next to cancer were collected from the Department of Pathology,Affiliated Hospital of Qingdao University from October 2017 to October 2018.None of the 100 patients were treated with radiation and chemotherapy or other treatments,and have been diagnosed with papillary thyroid carcinoma.Real-time PCR was used to detect the expression of lncRNA-PRSS3P2 and mRNA PRSS1,PRSS2 and PRSS3 in 100 cases of papillary thyroid carcinoma and adjacent tissues.The relationship between PRSS3P2 and Clinical characteristics was analyzed.And analyzed the expression correlation of PRSS1,PRSS2,PRSS3 and PRSS3P2.(2)Design and synthesize lncRNA-PRSS3P2full-length overexpression plasmid and interference shRNA.Transfection of thyroid papillary carcinoma BCPAP cells allows overexpression or inhibition of lncRNA-PRSS3P2 in cells.Transwell invasion and migration ability experiments were performed on transfected BCPAP cells.Changes in invasion and migration of BCPAP cells after expression or inhibition of lncRNA-PRSS3P2 were observed.Results:(1)The expressions of PRSS3P2 and PRSS1,PRSS2 and PRSS3 in 100 cases of papillary thyroid carcinoma were higher than those in adjacent normal tissues,and the difference was statistically significant(P < 0.05).However,PRSS1,PRSS2 and PRSS3 are not associated with the clinicopathologic features of PTC.The expression level of lncRNA-PRSS3P2 was associated with lymph node metastasis in patients,p<0.05(2)The lncRNA-PRSS3P2 full-length over expression plasmid and theinterfering shRNA was transfected into BCPAP cells,and the invasion and migration ability of BCPAP cell was determined.It was found that when lncRNA-PRSS3P2 was over expressed,its invasion and migration ability was enhanced,and the difference was statistically significant(P< 0.05).When the expression of lncRNA was inhibited,the invasion and migration ability was decreased,and the difference was statistically significant(P< 0.05).Conclusion: The expressions of PRSS3P2 and PRSS1,PRSS2 and PRSS3 in papillary thyroid carcinoma are higher than those in adjacent normal tissues,indicating that this group may be involved in the occurrence and development of papillary thyroid carcinoma.When over expressing and inhibiting lncRNA-PRSS3P2,it can affect the invasion and migration of papillary thyroid carcinoma BCPAP,indicating that lncRNA-PRSS3P2 may be involved in the invasion and metastasis of papillary thyroid carcinoma... |
