| Background : Huaiqi Huang Granule(Huaierjin)is a traditional Chinese medicine preparation widely used in respiratory,immune and kidney systems.Because of its safety and effectiveness,it has been accepted and welcomed by clinicians in various fields of Western medicine.In order to expand the market and delineate the field of neonates,enterprises established neonatal hypoxic ischemic encephalopathy(HIE)as the treatment object of Huaiqi Huang Granule.In order to grasp the first-hand information,the enterprise entrusted our department to start with animal experiments and observe its clinical effect and mechanism.Because the main clinical manifestations of HIE are alertness,spasm and convulsions,which belong to the category of convulsions in traditional Chinese medicine.Traditional Chinese medicine says that "all wind and dizziness belong to the liver" and "kidney governs the brain and generates marrow".Therefore,the pathogenesis of this disease in traditional Chinese medicine mostly belongs to deficiency of liver and kidney yin,obstruction of collaterals and wind,and nourishing yin,invigorating qi and activating blood circulation should be the main treatment of this disease.Based on this,the author chooses Huaiqi Huang Granule as reagent,establishes experimental animal model,observes the therapeutic effect of the preparation on HIE animal model,and preliminarily clarifies its mechanism and approach for HIE,thus proving the effectiveness and feasibility of the preparation in treating HIE,and providing strong evidence for its clinical application in neonates.Research objective : Based on the above research background,this study explores the mechanism of Huaiqi Huang Granule on neonatal ischemic-hypoxic encephalopathy by studying the literature and basic theory of HIE in traditional Chinese medicine,combining with the research results of modern western medicine on HIE,and according to the data of this animal model experiment,so as to provide reliable theoretical and experimental basis for clinical application of Huaiqi HuangGranule in treating HIE.Experimental method and detection index : In this experiment,Vannucci model was used as HIE model.Huaiqi Huang Granule was used to treat neonatal SD rats by gavage.To evaluate the cerebral ischemia and hypoxia and apoptosis of brain cells in neonatal rat HIE model by observing animal behavior and HE staining of pathological sections.Meanwhile,S100 B and Caspase-3 were determined by immunohistochemical staining SP,average optical density and enzyme-linked immunosorbent assay(Elisa).Experimental results:1.Behavioral observation: After successful modeling,all SD rats in the model group showed behavioral changes,mainly manifested as contralateral limb dyskinesia,difficulty in opening eyes and so on.SD rats in sham-operated group had no such changes.The early behavioral ability of SD rats in Huaiqi Huang group was similar to that in model group,and the later behavioral ability was less than that in earlier stage.2.HE staining observation results: At several specific time points after the completion of the model,the morphological expression of brain tissue cells of SD rats in sham-operated group was normal;the necrosis and nucleolysis of brain tissue cells of SD rats in model group were observed;compared with model group,the early changes of Huaiqi Huang group were similar to those of model group,and the later changes of brain tissue were better than those of model group,similar to those of sham-operated group.3.Immunohistochemical results:Compared with the concentration of S100 B and Caspase-3 in SD rats of sham operation group,the concentration of S100 B and Caspase-3 in SD rats of model group increased significantly at 2h,1d,3d and 7d(p < 0.01).The results indicated that the preparation of neonatal HIE animal model was successful;at 1d,3d and 7d after the establishment of model,Huaiqi Huang was obtained.The concentration of S100 B and Caspase-3 in the group was lower than that in the model group,with statistical significance(p < 0.05)or(p < 0.01),suggesting that Huaiqi Huang Granule could decrease the concentration of Caspase-3 and S100 B in the serum of neonatal HIE model rats.Research conclusion:1.Huaiqi Huang Granule is effective in repairing the brain tissue structure of SD rats with HIE.2.The content of Caspase-3 protease in brain tissue of neonatal HIE rats was decreased by intragastric administration of Huaiqi Huang Granule.3.The content of S100 B protein in the brain tissue of neonatal HIE rats was decreased by intragastric administration of Huaiqi Huang Granule.the above experimental studies show that Huaiqi Huang Granule can repair the brain tissue damage caused by cerebral ischemia and hypoxia,and reduce the apoptosis of brain nerve cells,so as to play a role in inhibiting the development of HIE. |
