| Objective: Renal interstitial fibrosis is a common pathological manifestation of many patients with chronic kidney disease to the end stage.Exploring effective methods to prevent renal interstitial fibrosis can delay the progress of chronic kidney disease.Therefore,the expression of Klotho protein in renal tissue with different degrees of fibrosis was detected,and the relationship between Klotho protein and renal interstitial fibrosis was further discussed.Methods: 69 cases of Ig A nephropathy and 20 cases of normal renal tissue adjacent to clear cell carcinoma were diagnosed as chronic kidney disease and renal tissue biopsy in the first affiliated Hospital of Medical College of Shihezi University during 2012.1-2018.1,and 20 patients with renal clear cell carcinoma and adjacent normal renal tissues.Hematoxylin-eosin,Masson staining technique to observe the pathological changes of renal tissue;2.Divided into normal control group(20 patients),fibrosis grade I group(25 patients),fibrosis grade II group(22 patients),fibrosis grade III group(22 patients);3.Immunohistochemical staining technique to detect the expression of Klotho protein and TGF-β factor in kidney tissue,Differences between groups were compared;The clinical indexes such as urinary β 2 microglobulin,serum creatinine and blood Ca,blood P were detected by biochemistry;correlation analysis between Klotho protein and renal interstitial fibrosis was performed.Results: 1.There was no significant difference in age,sex,weight and BMI among the four groups(P > 0.05).2.Among the biochemical indicators,Scr was statistically different among the four groups(p<0.05),and fibrosis group II(97.71±33.78)had higher Scr than the normal control group(75.41±13.05)and fibrosis group I.(82.64±12.16),the Scr of the fibrosis grade III group(118.66±31.89)was higher than that of the normal control group,the fibrosis grade I group,and the fibrosis grade II group;the Bun was statistically different among the four groups(p <0.05),the fibrosis grade I group(6.84±2.24)Bun was higher than the normal control group(4.63±1.24),the fibrosis grade II group(7.02±2.29)Bun was higher than the normal control group,and the fibrosis grade III group(7.64).±2.52)Bun was higher than the normal control group;blood Ca was statistically different among the four groups(p<0.05),and fibrosis II group(2.18±0.18)was lower than the normal control group(2.29±0.13)and Fibrosis grade I group(2.29±0.09),fibrosis grade III group(2.15±0.12)blood Ca was lower than normal control group and fibrosis grade I group;blood P was statistically different among the four groups(p<0.05).The fibrosis grade II group(1.30±0.17)had higher blood P than the normal control group(1.16±0.14)and fibrosis grade I group(1.18±0.12).The blood P of the grade III group(1.35±0.22)was higher than that of the normal control group,the fibrosis grade I group,and the fibrosis grade II group;the urine β2-MG was statistically different among the four groups(p<0.05),the fibrosis grade I group(0.78±0.53)urine β2-MG was higher than the normal control group(0.13±0.10),and the fibrosis grade II group(1.02±0.44)urine β2-MG was higher than the normal control group.The fibrosis grade III group(1.84±0.76)urine β2-MG was higher than the normal control group,the fibrosis grade I group,and the fibrosis grade II group.3.The expression of Klotho protein in renal tissues was statistically different among the four groups(p<0.05),and the Klotho protein in the fibrosis group II(2.68±2.01)was lower than that in the fibrosis group I(4.00±2.27).(1.55±0.80)Klotho protein was lower than the normal control group(3.50±1.70),fibrosis grade I group and fibrosis grade II group;the expression of TGF-β factor was statistically different among the four groups(p<0.05).The fibrosis grade I group(1.72±1.17)had higher TGF-β factor than the normal control group(1.00±0.97),and the fibrosis grade II group(3.00±2.20)had higher TGF-β factor than the normal control group and fibrosis I.In the grade group,the fibrosis grade III group(4.65±1.97)had higher TGF-β factors than the normal control group,the fibrosis grade I group,and the fibrosis grade II group.4.Klotho protein level in renal tissue was positively correlated with blood Ca(P<0.01),and TGF-β factor,renal interstitial fibrosis renal interstitial fibrosis score,TGF-β,Scr,urine β2-MG,age,blood P Negative correlation(P<0.01).There was no correlation between Klotho protein expression and Bun(P>0.05).Conclusion: With the increase of renal interstitial fibrosis,the expression of Klotho protein is decreasing.and Klotho protein is associated with renal interstitial fibrosis. |
