Relationship Between Matrix Metalloproteinase-3 Polymorphism And The Capillary Bronchitis With RSV Infection

Objective:To investigate the possible association of matrix metall oproteinase-3 gene polymorphism with the susceptibility and severity of RSV infection.We study the distribution among different groups of genotype and alleles of genetic locus rs522616 and rs 679620 in MMP-3.To assess the value of single nucleic acid polymorphism of genetic lo cus rs522616 and rs679620 in predicting the severity and morbidity of RSV infection.Methods:We accept 145 children between 2-24 months as case group,w ho are RSV positive and have been diagnosed with capillary bronchitis refer to the eighth edition of practical pediatrics during January 2017 to December 2018.Chose 124 RSV negative healthy children as the c ontrol group.And all children are divided into allergic constitution group(54)and non-allergic constitution group(215).Case group is divided into mild group(122)and sever group(23).Recording allergy hi story,cardiopulmoanary disease history,treatment,comlications,hospita l stays of all children.All children have excepted premature,congenit al heart disease,pulmonary dysplasia and so on.Sampling 1 ml blood of all children as specimen.All parents have signed the informed consen t.Primer are projected with Primer-Blast software.Blood samples are e xtracted,amplified,electrophoresed and purified.Than complete the seq uencing with ABI.3730XL sequencer.Find the information of genetic loc us of rs522616 and rs679620 in NCBI.Analyze the result of sequencing with Mutation Surveyor,then get the genotype of rs522616 and rs679620.Result:There are just A and G alleles on the rs522616 and rs679620on MMP-3 gene.1.The genotype frequencies of rs522616 in case group are AA(35.9%),A G(49.0%)and GG(15.1%),and the allele frequencies are A(60.3%)and G(39.7%).There are no significant differences of the frequencies betw een case group and control group(P>0.05)or between mild group and s ever group(P>0.05).The frequency of AA-genotype on rs679620 in aller gic constitution group is higher than non-allergic constitution group(X~2=8.679,P<0.05).2.The genotype frequencies of rs679620 in case group are AA(36.9%),A G(55.9%)and GG(35.1±7.9%),and the allele frequencies are A(36.9%)a nd G(63.1%).The A-allele frequency in case group is higher than contr ol group(X~2=4.127,P<0.05).And the AG-genotype frequency in sever gro up is higher than mild group(X~2=7.931,P<0.05).There are no significa nt differences of frequencies between allergic constitution group an d non-allergic constitution group(P>0.05).2.Conclusions:A-allele on rs679620 on MMP-3 gene is related to the susceptibility of capillary bronchitis with RSV infection in childre n in Hangzhou.And AG-genotype on rs679620 on MMP-3 gene is related to the severity of capillary bronchitis with RSV infection.Gene polymor phism of rs522616 is not related to the susceptibility and severity o f capillary bronchitis with RSV infection.But AA-genotype on rs522616is related to the allergic constitution.
