Objective:1 Through the model of Ovarian Hyperstimulation in the rat,discuss the effectiveness of reinforcing kidney and activating blood circulation through the LIF/STAT3 signaling pathway improving the endometrial receptivity.2 Through the model of Ovarian Hyperstimulation in the pregnant rat,discuss the effectiveness of reinforcing kidney and activating blood circulation through the LIF/STAT3 signaling pathway improving the endometrial receptivity of the pregnant rats.The experiment demonstrated that the reinforcing kidney and activating blood circulation can improve the expression of endometrial receptivity factors,discussed the effect target of the reinforcing kidney and activating blood circulation so as to clarify the mechanism of traditional Chinese medicine improving the endometrial receptivity and increasing the chances of pregnancy and provide scientific basis for further applicaton in ART.3 Through the clinical researches of the effectiveness of reinforcing kidney and activating blood circulation acting on ovulated PCOS complicated with infertility of kidney deficiency and blood stasis syndrome,proven that the reinforcing kidney and activating blood circulation can increase the thickness and improve the types of the endometria and increase the pregnancy rate.Methods:1 Using GnRHa+PMSG+HCG to establish the model of Ovarian Hyperstimulation in the rat,through HE staining,observe the thickness of theendometria through the light microscope;Through IHC,detect endometrial MVD,integrin beta 3 and LIF,STAT3,p-STAT3.2 Using GnRHa+PMSG+HCG to establish the model of Ovarian Hyperstimulation in the rat and mating the male rats and the female rats in proportion of 2:1 after injection HCG,the rats with vaginal plug is the symbol of pregnancy.Through IHC,detect endometrial MVD,integrin beta 3 and LIF,STAT3,p-STAT3.3 Through the clinical research,we employ the plan of CC+HCG to ovulate for the patients of PCOS complicated with infertility.Examine the endometrial thickness and types in the follicular maturation day by Transvaginal color Doppler sonography and compare the pregnancy rate simultaneously.Results:1 HE staining results of endometrial thickness of the unfertilized ratsHE staining manifested that the endometrial thickness of the normal group and the herbal group in metestrus is significantly higher than the model group(P<0.05).The difference was not statistically significant between the normal group and herbal group(P>0.05).The endometrial thickness of the normal group and the herbal group in anestrus is significantly higher than the model group(P<0.05).The difference was not statistically significant between the normal group and herbal group(P>0.05).The endometrial thickness of the herbal group in proestrus and estrus significantly increased statistically than the normal group and the model group,and the endometrial thickness of the model group significantly decreased statistically than other groups(P<0.05).2 IHC results of endometrial MVD,integrin beta 3,LIF,STAT3 and p-STAT3 of the unfertilized ratsImmunohistochemical method manifested that compared with the model group,the expression of endometrial receptivity related factors including MVD,integrin beta 3,LIF,STAT3 and p-STAT3 of the normal group is significantly higher than the model group(P<0.05).Compared with the modelgroup,the endometrial factors above significantly increased statistically in the herbal group(P<0.05).3 IHC results of endometrial MVD,integrin beta 3,LIF,STAT3 and p-STAT3 of the pregnant ratsImmunohistochemical method manifested that compared with the model group,the expression of endometrial receptivity related factors including MVD,integrin beta 3,LIF,STAT3 and p-STAT3 is significantly higher than the model group(P<0.05).Compared with the model group,the endometrial factors above significantly increased statistically in the herbal group(P<0.05).4 The endometrial thickness in the follicular maturation dayThe clinical study indicated that compared with the herbal group,the endometrial thickness in the follicular maturation day is statistically higher than the normal group(P<0.05).5 The endometrial type in the follicular maturation dayThe endometrial type A accounting for 60% in the herbal group is statistically higher than 37% of the normal group(P<0.05);The endometrial type B accounting for 37% in the herbal group is statistically lower than 47%of the normal group(P<0.05);The endometrial type C accounting for 3% in the herbal group is statistically lower than 16% of the normal group(P<0.05).6 Comparison of pregnancy rateThe pregnancy rate of the herbal group is statistically higher than the normal group(P<0.05).Conclusions:1 Reinforcing kidney and activating blood circulation herbs may regulate the angiogenesis through adjusting the expression of LIF/STAT3 related factors to increase blood supply of uterine,improve the uterine microenvironment and enhance the endometrial receptivity in later period.Reinforcing kidney and activating blood circulation herbs can improve the endometrial receptivity through the LIF/STAT3 signaling pathway.2 Reinforcing kidney and activating blood circulation herbs may regulate the angiogenesis through adjusting the expression of LIF/STAT3 related factors to improve the uterine microenvironment and enhance the endometrial receptivity,which is good for the maintenance of pregnancy.3 Reinforcing kidney and activating blood circulation can increase the endometrial thickness,improve the endometrial types and raise the pregnancy rate in the ovulated PCOS complicated with infertility.
