Objective The studie discusses the genetic association between SNP rs 2333227 in MPO gene and the myeloperoxidase level of plasma with KD susceptibility and coronary artery lesion in Inner Mongolian children inadequate.Methods We selected 35 children with Kawasaki Disease as the case group who were treated in Pediatrics Department of Inner Mongolia People’s Hospital from October 2017 to October 2018.The control group consisted of 18 children who had health check-up at the same time.KD was divided into two subgroups according to the presence or absence of concurrent CAL: CAL and NCAL.Strictly abide by the aseptic operation principle,2 ml of venous blood were collected.1.MPO levels in the plasma were measured using the ELISA technique.Through the comparative study of KD Group and Control Group as well as CAL and NCAL subgroups MPO levels in the plasma reached the corresponding conclusion.2.First,using a kit to extract the gene.Second,using polymerase chain reaction to amplify the target fragment.Last,the SNP rs2333227 of MPO gene in KD Group and Control Group as well as CAL and NCAL subgroups was analyzed by sequencing.Results1.In KD group,35 cases were aged from 5 months to 7 years,the average age was 1.87±0.54 years.There were 25 males and 10 females(2.5:1).The Control Group(N = 18),aged from 5 months to 7 years,had an average age of 2.56±0.86 years.There were 13 males and 5females,and the ratio was 2.6:1.In Cal Group,there were 9 cases(mean age 1.72 ± 0.56years),8 male and 1 female,male to female ratio was 8:1.In NCAL group,there were 26patients(mean age 1.92 ± 0.69 years),17 males and 9 females,male to female ratio was 1.8:1.In Cal Group,6 patients had unilateral coronary artery dilatation,2 patients had bilateral coronary artery dilatation and 1 patient had coronary artery aneurysm.2.In the control group,the MPO concentration was 83.688±7.301(ng / L).In KD Group,the MPO concentration was 88.587±8.165(ng / L);The concentration of MPO in KD group was significantly different from that in control group(t=1.686,p=0.032).The MPO level of plasma in CAL group was 93.974±7.555(ng / L);the MPO level of plasma in NCAL group was 86.721±7.638(ng / L).MPO level of plasma in CAL group was significantly higher thanthat in NCAL group(t=1.761,p=0.027).3.The genotype frequencies(GG,GA,AA)of MPO gene locus(RS2333227)in KD Group and Control Group were statistically significant(c2=4.176,p =0.041);There were significant differences in allele frequency(c2=4.706,p=0.030),The G Allele is a risk factor.There were significant differences in genotype between CAL group and NCAL group(c2=4.137,p=0.042),as well as in allele frequency(c2=4.117,p=0.043).Conclusion1.The level of MPO in KD Children was higher than that in NCAL children,and MPO in Cal Group was higher than that in NCAL group.2.The MPO gene of rs2333227 site is associated with KD,and the G allele is a predisposing factor,which may be associated with coronary artery injury in Kawasaki Disease.
