Objectives Myeloperoxidase (MPO), a leukocyte enzyme that promotes oxidation of lipoproteins in atheroma, has been proposed as a possible mediator of atherosclerosis. There is a functional polymorphism 463 bp upstream of the enzyme. A lot of results have been reported overseas on the association between this polymorphism and coronary artery disease(CAD). There is few civil relative research. In this study, the importance of MPO polymorphism was further evaluated in Chinese.Methods To investigate the relation between MPO polymorphism and CAD, we conducted a case-control study of 105 case patients with myocardial infarction(MI) or unstable angina(UA) and 105 controls without known heart disease. PCR-RFLP method was used to determine genotypes in all these subjects enrolled in this study. The serum MPO activity was measured by colorimetric method. And at the same time, we measured the serum levels of TC, TG, LDL, HDL.Results The frequency of alleles differ significantly between patients groups and controls (X~2= 10.142, P<0.05) . When analyzing the clinical material, we found there is not relationship between genotypes of MPO and smoking , hypertension , diabete and hyperlipidemia. Our study indicated that allele G of the MPO gene was more frequent in cases with CAD. Patients with the GG genotype had an increased risk of CAD (odds ratio8.3, 95% confidence interval 2.046-13.443); however patients with the AA genotype had a decreased risk of CAD.
