| Objective:To observe the effects of brain tissue and immune function with Concussion Rats Treated by brain Vibration Therapy,To expounds the mechanism and principle of the treatment concussion by Mongolian medicine brain Vibration Therapy.Methods:The 80 SPF-grade SD rats(male,weighing 250±20g)were randomly divided into ten groups:control grouP,model 4 days,model 7 days,model 10 days,westerm medicine 4 days,western medicine 7 days,westerm medicine 10 days,2 brain vibration therapy group,3 brain vibration therapy group,4 brain vibration therapy group,a total of 10 groups.Animal model of brain concussion in Rats was duplicated with a metallic pendulum-striker concussive for the other 9 groups except the control group.The control group did not under go any treatment.Models of 4 days,7 days of model,and 10 days of model did not undergo any treatment when the brain concussion model was replicated.Western medicine 4 days group,western medicine 7 days grouP,and western medicine 10 days group replicated the concussion model respectively.Nimodipine tablets(5mg/kg.d)were administered intragastrically for 4 days,7 days,and 10 days.The 2 brain vibration therapy group received brain vibration therapy on the same day and on the fourth day after the concussion model was replicated,the 3 brain vibration therapy group received brain vibration therapy on the same day and on the fourth and seventh day after the concussion model was replicated,the 4 brain vibration therapy groups performed brain vibration therapy on the same day,the fourth,seventh and tenth days of the brain concussion model.On the fourth,seventh and tenth day of the experiment,Nissl staining was used to show the changes of hippocampus neurons in concussion rats.The content of IL-1β,IL-6 and IL-8 and TNF-α in serum were determined by ELISA.Western Blot method was used to detect the changes of 5 HT and Ach in hippocampus,cortex and hypothalamus of rats.All data were processed using SPSS 22.0 statistical software.Result:1.Pathological examination:In the control group,neurons were larger in volume,regular in shape,the cell layers had tight order,elear in structure and the number of Nissl body was rich;In the model group,the volume of nerve cells was larger,the arrangement was disorder,the shape of some neurons was irregular,and the number of Nissl body decreased;The western medicine group was similar to the model group;In the brain vibration therapy group,most of the neurons were intact in shape,the cells were arranged orderly and dense,and some of the content of Nissl body was increased.Although,compared with the control group,the number of neurons and the content of Nissl body were different,the morphology and structure of the neurons were close to that of the control group.Compared with the model group and western medicine group,there was significant improvement in all aspects.2.The ELISA method detects:The contents of IL-1β,IL-6 and IL-8,TNF-α in each model group were significantly higher than those in the control group(P<0.05).The content of IL-1β in the 4-day western medicine group,compared woth the model group,was significantly reduction.The content of IL-6 was significantly decreased in the western medicine 4-day group,in the 7-day group and 2 brain vibration therapy group.And the content of IL-8 in the western medicine 4-day group and the western medicine 10-day group was significantly decreased.Otherwise the content of TNF-αin the 4-day,7-day group and 2 brain vibration therapy group was significantly lower than that in the model group(P<0.05).In addition,the content of IL-6,IL-8,TNF-α in each treatment group was lower than that in model group.3.The Western Blot method detects:Compared with the normal group,the content of 5-HT in hippocampus increased significantly in the 4-day model group and 7-day model group,and in the hypothalamus and cortex of the model-4-day group the content of 5-HT increased obviously while the content of Ach were decreased significantly,and there was statistical significance(P<0.05).Compared with the model group,not only the content of 5 HT in the cortex of western medicine 4 days and 2 brain vibration therapy group was significantly decreased,but also the content of 5 HT in hypothalamus was significantly decreased in the 2 brain vibration therapy group.In addition,the content of Ach was increased significantly in the cortex of 2 brain vibration therapy group and hypothalamus of western medicine 4 days(P<0.05).The contents of neurotransmitters in the brain tissue of the 10-day model group,10-day western medicine group and 4 brain vibration therapy group were similar to those of the control group,especially in the treatment group.Conclusion:1.Pathological examination showed that in the model group,not only the number of Nissl bodies in hippocampus neurons decreased but also the function of neuron synthetic protein was decreased.However there was no obvious damage of hippocampus neurons after brain vibration therapy,which suggested that the treatment could improve the functional state and cell morphology of neurons,and reduce or delay the loss of neurons.2.The content of IL-1β,IL-6 and IL-8,TNF-α was increased by brain concussion.Brain vibration therapy could down-regulate the content of IL-1β,IL-6,IL-8 and TNF-α,suggesting that brain vibration therapy has a regulatory effect on immune function.3.Mongolian medicine brain vibration therapy can also regulate the content of neurotransmitters in the brain tissue of rats,so as to reduce the brain damage,improve and regulate the memory of learming,cognition and motor function.4.Mongolian medicine brain vibration therapy mainly to regulate the physique,can regulate the disorders of "the three GenSu",so that the affected brain and other viscera organs reset,recovery of brain injury and other effects on the brain concussion rats play a good role.And has the operation simple,the safety,the effective,the less toxicity side effect and so on the characteristic and the superiority.It is worth popularizing and applying widely. |
