Notopterygii Rhizoma ET Radix(Qianghuo)is from the dried roots and rhizomes of Notopterygium incisum Ting ex H.T.Chang and Notopterygium forbesii H.de Boiss.It contains a large amount of compounds,such as volatile oils,coumarin,sugar,amino acid,organic acid,plant stero,flavone and so on.It has extremely high medicinal value,and is commonly used medicinal materials for Tibetan,Yi,Qiang and other ethnic groups.Its property is Pungent,bitter and warm and acts on bladder and kidney meridian.And it has the functions of dispersing cold,dispelling wind,dehumidifying and relieving pain.In recent years,the Habitat of No.incisum and No.forbesii has suffered serious damage,and the resources of wild No.incisum and No.forbesii are nearly endangered because of over-excavation.Compared with roots and rhizomes,the stems and leaves of Qianghuo are cheap and easily available and contain similar chemical components of rhizomes.However,there is no systematic study on the stems and leaves of No.incisum and No.forbesii.In this study,the morphological and microscopic characteristics of the stems and leaves of No.incisum and No.forbesii were identified.The chemical composition,fingerprint and antioxidant activity in vitro were compared and studied.These results of the study will provide scientific basis for the quality evaluation,resource development and utilizations of the stems and leaves of No.incisum and No.forbesii.The main research contents are as follows:(1)Morphological and microscopic identification of stems and leaves of No.incisum and No.forbesii were identified.(2)Thin layer chromatography(TLC)was used to study the stems and leaves of No.incisum and No.forbesii,compared with notopterol and isoimperatorin as control.(3)Compare and analyze the main chemical components of stems and leaves of No.incisum and No.forbesii by ultra-high performance liquid chromatography.Determine the contents of four compounds including notopterol,isoimperatorin,bergapten and diosmin.Analyze samples from 22 producing areas,and establish fingerprints of stems and leaves of No.incisum and No.forbesii,to determine a common characteristic peak.Comparing similarity between samples from different producing areas by similarity analysis and cluster analysis,it provided basis for qualitative identification of stems and leaves of No.incisum and No.forbesii.(4)Taking rutin as reference,total of flavonoids of stems and leaves of No.incisum and No.forbesii were determined by UV-spectrophotometry.And the content of total flavonoids decline in sequence from No.incisum leaves,No.incisum stems,No.forbesii leaves to No.forbesii stems.(5)The scavenging ability of stems and leaves extracts of No.incisum and No.forbesii were determined by DPPH and ABTS+ radicals and the reducing ability of Fe3+ models.And the order of antioxidant capacity: No.incisum leaves > No.incisum stems > No.forbesii leaves > No.forbesii stems.(6)UHPLC QTOF-MS/MS method to quickly identify chemical components in the stems and leaves of No.incisum and No.forbesii were established.And mass spectrometry conditions : Column: Wters Acquity HSS-T3(1.8 ?m,2.1 × 100 mm);mobile phase: acetonitrile(B)-0.1% formic acid water(A);gradient elution: 10-20% B(0-10min),20-30%B(10-20min),30-40%B(20-30min),40-57B%(30-33min),57-90 %B(33-43min),90-90%B(43-48min),90-10%B(48-49min),10-10%B(49-54min).Detection wavelength 330 nm,flow rate 0.3 m L/min,column temperature 30 ?,injection volume 1 ?L.Mass spectrometry conditions: The electrospray ionization(ESI)ion source collects data in positive and negative ion modes,with a mass scanning range of m/z 100~1000,ion source parameters of 3200 V capillary voltage,40 k V cone voltage,ion source temperature of 100 ?,Desolvent gas temperature of 350 ?,Desolvent gas(N2)volume flow rate of 700 L/h,and collision energy(CE)of 30 ~ 45 V.A total of 36 compounds were detected in the steam of No.incisum and No.forbesii.,and 32 chemical components were identified,the remaining four components were unknown.A total of 41 compounds were detected in the leaves of No.incisum and No.forbesii.,,and 38 chemical components were identified,the remaining three components were unknown by molecular ion peak,fragment ion peak and literature comparison.
