Objective: To investigate the expression profiles of MAPK signaling molecule m RNA and protein in leiomyoma and uterine myometrium.Methods: A total of 20 leiomyoma patients who underwent laparoscopy were recruited in this study.The m RNA and protein expressions of RAS、RAF、MEK、ERK in leiomyoma and myometrium of uterine leiomyoma patients were detected,the myometrium as the control group.Results: The m RNA expression of RAS、RAF、MEK、ERK in leiomyoma was higher than that in myometrium(p<0.05).The protein expression of RAS、RAF、MEK、ERK in leiomyoma was higher than that in myometrium(p < 0.05).The protein expression of p-MEK1/2、p-ERK1/2 in leiomyoma was significantly higher than that in myometrium(P<0.01).Pearson correlation analysis showed that the expression of RAS、ERK、p-ERK was positively correlated with menstrual flow(P<0.05).Spearman correlation analysis showed that the expression of RAF was positively correlated with uterine volume and leiomyoma volume(P<0.05)Conclusions: The expression of MAPK signaling pathway molecule in leiomyoma is higher than that in the adjacent myometrium,and the protein phosphorylation level of MEK1/2 and ERK1/2 was significantly enhanced,suggesting that MAPK signaling pathway was activated in leiomyoma.Menstrual flow,uterine volume,leiomyoma volume and MAPK signal pathway expression was positively correlated,suggesting that MAPK signal pathway play an important role in the progress of leiomyoma.
