Lysimachia Christinae Hance is a traditional Chinese medicine.It can be used in the treatment of lithiasis,urinary system infections and other diseases.In this study,the constituents of Lysimachia Christinae Hance were analyzed and the extraction of quercetin flavones by using deep eutectic solvents was investigated.The following parts are included:Firstly,an UPLC method for the analysis of the extract of Lysimachia Christinae Hance was established.The key chromatographic conditions including the chromatographic column,mobile phase system,column temperature and flow rate were optimized.Under the optimized condition,the method validation was accomplished.The results of method validation demonstrated the developed method possessing desirable precision,reproducibility and accuracy.Secondly,the constituents of Lysimachia Christinae Hance extract were identified by ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry(UPLC-QTOF-MS).The 45 chromatographic peaks were detected,and 30 of them were tentatively assigned and identified.These constituents were mainly phenolic acids,flavonoids and fatty acids.Finally,the quercetin and flavones from Lysimachia Christinae Hance were extracted by deep eutectic solvents.Eleven kinds of deep eutectic solvent were successfully prepared,which were all present as liquid at room temperature.All of the deep eutectic solvents were applied in the extraction and 50 % methanol was used as extraction reference solution for comparison.The choline chloride-carboxylic acid system was found to be the good extraction solvent for quercetin flavones.Extraction conditions including solvents selection,extraction time and extraction temperature were optimized by single factor experiments to obtain a good extraction condition.
