Objective: the purpose of this experiment was to investigate the effect of the combination of Chinese herbal medicine Lysimachia christinae Hance and Lithium Carbonate on the intestinal flora of calcium oxalate rats using a high-throughput sequencing method.Methods: 48 adult male SD rats were randomly divided into 8 groups(Z,M,JQC,L,JLD,JLZ,JLG,PC).The rats were fed by 1% Glycol solution free water and 2% Ammonium Chloride 2ml gavage to build calcium oxalate kidney stone rat model.Specimens were collected from rats in each group after 56 days of intervention under the same conditions.The whole genome of prokaryotic organisms was extracted from the fecal specimens of each group,and the colony structure was analyzed by 16 S rDNA V3/V4 region gene sequencing.The left kidney tissue was observed under the microscope for histopathological changes and crystalline deposits.To explore the effect of pharmacological intervention on changes in intestinal flora of rats with calcium oxalate kidney stones.Results:(1)Results of paraffin sections of left kidney: Compared with group Z,group M showed large and widespread piles of calcium oxalate crystals deposition,which were connected into sheets;the crystals formed in group JQC,JLD,JLZ and PC were less than those in group M;the performance within group L and group JLG was differ from one slice to another.(2)Results of high-throughput sequencing : The relative abundance and species diversity of gut flora in the drug treatment group were higher than those in group Z and M.The relative abundance and species diversity of gut flora in JLZ group and PC group were more unanimous with the other.(3)At the phylum level,8 bacterial phylum were detected in all groups.Among all of them,Firmicutes,Bacteroidetes,Actinobacteria,Proteobacteria and Tenericutes were account for more than 99%.Group JLG were significantly different at the level of Tenericutes,Actinobacteria,Bacteroidetes,Firmicutes.For the remaining groups,there were little difference in them.(4)At the genus level,Lactobacillus is the most dominant genus of all groups,however,the relative abundance of the flora varies depending on the lithogenic agent and the drug.Conclusion: The intestinal microflora play an important role in maintaining the health of the body.The dysfunction of its structure and quantity are related to many diseases,which including urinary stone disease.Lysimachia christinae Hance combined with Lithium Carbonate may affect the formation and deposition of calcium oxalate crystals in rat kidney by influencing the composition and quantity of intestinal flora.Lysimachia christinae Hance,Lithium carbonate,and combined use of these two drugs all have the ability to affect the number and structure of intestinal microflora,and different concentrations of Lithium Carbonate have different effects on intestinal microflora.
