| Objective: To observe the effect of moxibustion at stomach meridian acupoints on the repair of gastric mucosal injury in rats with peptic ulcer(PU).To explore the mechanism of moxibustion at stomach meridian points on PU rats from the aspects of histopathology,molecular biology and genomics.Methods: 24 male SD rats were randomly divided into: control group,peptic ulcer group(PU group),moxibustion on the stomach meridian acupoints(ST group)and moxibustion on the gallbladder meridian acupoints(GB group).PU rats model were established by intragastric administration of 70% ethanol solution.All rats were be moxibustion at stomach meridian acupoints(“Liangmen”,“Zusanli”)and gallbladder meridian acupoints(“Riyue”,“Yanglingquan”)after establishment of the model successfully.Gastric ulcer index were measured after intervention.In addition,the gastric mucosa and feces of rats in each group were collected.The expression of Gastrin(GAS)and epidermal growth factor(EGF)in gastric mucosa were detected by quantitative real-time PCR(qPCR).16 S rDNA gene sequencing was used to detect the relative expression and species of bacteria in feces.Result:(1)Histopathological observation showed that the cells of gastric mucosa in the control group were arranged neatly and the gastric mucosa was intact.The gastric mucosa in PU group was damaged obviously and the cells were disordered.Compared with the PU group,the injury of gastric mucosa was obviously improved in ST group,and the arrangement of gastric mucosal cells was neater than that in PU group.In the GB group,gastric mucosal injury still existed,and the arrangement of gastric mucosal cells was more orderly than that in the PU group.However,there are still a few inflammatory cells in the gastric mucosa.(2)The index of gastric ulcer showed that: compared with the PU group,the gastric ulcer index of the ST group was lower significantly(P<0.05),the gastric ulcer index of the GB group was decreased,but there was no difference between the GB group and the PU group(P >0.05).(3)The results of qPCR in gastric mucosa showed that: compared with the PU group,the expression of GAS and EGF in gastric mucosa of control group and ST group were lower significantly(P<0.05).The expression of GAS and EGF in GB group was increased,but there was no difference compared with the PU group significantly(P>0.05).(4)The results of 16 S rDNA gene sequencing showed that the relative content of bacteria in control group was lower than that in PU group,ST group and GB group significantly(P<0.05).The relative content of bacteria in ST group was higher than that in PU group and GB group significantly(P<0.05).There was no difference in the relative content of bacteria between PU group and GB group significantly(P>0.05).In the control group,the relative expression of oscillospira,prevotellaceae and tenericutes was higher.In the PU group,the relative expression of bacteroidia,bacteroidales and ruminococcaceae was higher.In the ST group,the relative expression of alphaproteobacteria,actinomycetales and bacillales was higher.In GB group,the relative expressions of firmicutes,clostridia and lachnospiraceae were higher.Conclusion: Moxibustion at stomach meridian acupoints can repair pathological lesions of gastric mucosa in rats with peptic ulcer significantly.Moxibustion at stomach meridian acupoints can regulate the imbalance of GAS and EGF factors in rat gastric mucosa with peptic ulcer,and promote the reproduction of gastrointestinal flora to repair mucosal lesions. |
