Objective To investigate the changes of lymphocyte subsets and serum levels of related cytokines in patients with immune thrombocytopenia(ITP)before and after treatment with recombinant thrombopoietin(TPO):1.Shift of Th1/Th2 and Treg/Th17 balance in patients with chronic ITP,thereby improving the understanding of pathogenesis of patients with chronic ITP;2.To study the number and function of Th1,Th2,Th17 and Treg of T lymphocyte subsets,as well as the influence of cytokines secreted by lymphocytes,such as IL4?IL-6?IL-10?IL-1l and IL-17a,so as to provide basis for their treatment of immune-related thrombocytopenia;Methods From January 2018 to December 2018,10 patients with ITP diagnosed by flow cytometry were selected from the hematology department of wuhan central hospital,The percentage of lymphocyte subsets in peripheral blood of 10 patients with ITPtreated before and after TPO were detected by flow cytometry,including Th1?Th2?Th17?Treg,ELISA was applied to detect the level of Serum IL-2,IL-4,IL-6,IL-10,TNF-?,IFN-?,and IL-17A of 10 patients with ITP treated before and after TPO.Results 1.The percentage of CD3~+CD4~+CD8~-IL4~+cells and the ratio of CD3~+CD4~+ CD8~-IL~-17a~+in ITP group before TPO treatment were significantly higher than that after TPO treatment[(14.75±2.16)%vs(7.12±1.70)%,(6.70±0.69)%vs(3.80±0.42)%,all P<0.05].After TPO to treat ITP group of CD4~+CD25~+CD127~-ratio is higher than before treatment(1.18±0.36)%vs(7.46±0.87)%,P values<0.05.2.TPO ITP group after treatment serum IL-4,IL-6,IL-17a group were significantly lower than before treatment ITP,P values<0.05.Conclusion In ITP patients,there is an imbalance in lymphocyte subsets and abnormal cytokine levels,which can be corrected by TPO.
