| Purpose:The main characteristics of osteoporosis are bone loss caused by imbalance in bone resorption and bone formation.Postmenopausal osteoporosis often occurs insidiously in postmenopausal women.Brittle fracture is the most common clinical complication,which seriously endangers human health and quality of life.OPG/RANK/RANKL signaling pathway is an important pathway to regulate bone metabolism and balance,which is mainly involved in bone reconstruction by regulating osteoclast activation and promoting bone resorption.Probiotics are a kind of human intestinal flora.Recent studies have shown that probiotics can improve bone metabolism by regulating hormone and immune function.However,the content of probiotics decreases with age,which is consistent with the decrease of bone mass.However,the mechanism by which probiotics regulate bone metabolism is not well understood.Therefore,the study on the expression of OPG and RANKL in SD rat model of ovariectomy osteoporosis induced by probiotics is of great significance for further exploring the mechanism of probiotics in regulating bone metabolism and improving the bone mass of the elderly.Method:Fourty five 12 weeks old SD female rats were randomly were evenly divided into control group(Sham operation,Sham),ovariotomy group(Ovariectomy,the OVX),ovarian resection and probiotic preparations intervention group(OVX+Probiotics),each group of 15,in addition to the Sham group ovaries removed around the same size of adipose tissue,the OVX+Probiotics group and the OVX rats were performed bilateral ovaries removed.Eight weeks after the surgery,each group randomly,2 rats dual-energy X-ray absorptiometry measurement,determine after the success of the building,the OVX+Probiotics group feed gavage quad living bacterium equivalent dose concentration solution,Sham group and the OVX group was given distilled water filling and feed volume,for four consecutive weeks,put to death all the rats,Micro-CT measurement of three groups of rats femur lower Tb.N,Tb.Th,Tb.Sp,BV/TV,serum BGP and ALP,Ca level,OPG and ARNKL protein expression.Results:Compared with the Sham group,compared with the,the OVX+Probiotics group and the OVX rats femur lower Tb.N,Tb.Th,BV/TV value decreased significantly,Tb.Sp increased significantly,serum BGP and ALP,Ca levels were significantly lower(P<0.05),the OVX+Probiotics group compared with OVX group,the rat femur lower Tb.N,Tb.Th,BV/TV value,Tb.Sp,reducing RANKL protein expression and serum BGP and ALP,Ca levels,OPG protein expression were higher,The difference was statistically significant(P<0.05),after 4 weeks intragastric administration.Conclusions:1.BMD and BMC results showed that the ovariectomized osteoporosis model of SD rats was successfully established;2.Compared with OVX group,the values of Tb.N,Tb.3.Serum biochemical indexes of BGP,ALP and Ca were detected,and the results showed that probiotics(tetraphyllum tablets)could improve bone metabolism,inhibit bone resorption,promote bone formation and reduce bone loss.4.Expression of OPG and RANKL proteins.The results showed that probiotics(tetraploid live tablets)had an effect on the OPG/RANKL/RANK axis system,and thus played an anti-op role,providing a new research direction for the prevention and treatment of postmenopausal osteoporosis. |
