| Objective: Multiple myeloma SET domain(MMSET),also known as WHSC1 or NSD2,is a histone methyltransferase,which is closely related to physiological processes such as human embryonic development.Several studies show that MMSET is overexpressed in a variety of cancers and associated with poor prognosis of patients.In this study,we aimed to observe the differential expression of MMSET gene in ovarian cancer and the effect of gene silencing on the biological behavior of ovarian cancer cells.Methods: RT-PCR and Western Blot was performed to determine the differential expression of MMSET in ovarian cancer,normal ovarian tissues.Then the correlation between the expression of MMSET and the clinicopathological parameters of ovarian cancer patients was analyzed.The effects of MMSET gene silencing on the migration ability,invasion ability and p-AKT expression of ovarian cancer cells were detected by scratching,Transwell and Western Blot experiment.Results: The expression level of MMSET in ovarian cancer tissues was significantly higher than that in normal ovarian tissue and correlated with lymph node metastasis,abdominal water volume and CA125 level(P < 0.05),but irrelevanted with age,FIGO staging and tumor size.The nick distance of the experimental group was significantly wider than the control group(P<0.05).The number of cells in the control group was significantly higher(P<0.05).The expression of p-AKT in the experimental group was significantly lower than the control group(P<0.05).Conclusions: MMSET is highly expressed in ovarian cancer and significantly affected the migration and invasion ability of ovarian cancer cells.It is suggested that MMSET gene may be a potential target for the prevention and treatment of high grade serous ovarian cancer. |
