Objective: To study the effects of the "dry-heat-xin" national drug Fomes officinialis Ames on immune function and the effects of serum and urine metabolites on metabolomics based on nuclear magnetic resonance(NMR).Find the possible biomarkers and metabolic pathways related to cold-damp syndrome,verify drug targets related to "divergence" efficacy,comprehensively evaluate Fomes officinialis Ames on the body’s metabolic network,and reveal its chemistry and Network relationships between biological foundations.Methods: Forty SPF SD rats were placed in an artificial climate chamber in a cold and wet environment for 21 consecutive days.Through the observation of the rats’ general condition and the detection of endogenous indicators,it was verified that the rat model of cold-damp syndrome was successfully seted up.The material was obtained 14 days after intragastric administration of Fomes officinialis Ames.Evaluate the effects of Fomes officinialis Ames on the immune function of rats with cold-damp syndrome were determined by using the Elisa test kit and the organ coefficient of immune organs.Based NMR metabolomics were to find differential Small molecule compounds in serum and urine between rats in the cold-damp syndrome group and the administration groups.Principal component analysis(PCA)and partial least squares-discriminant analysis method(PLS-DA)was used to investigate the effects of Fomes officinialis Ames on metabolites and it’s pathways in rats with cold-damp syndrome.Results: After dosing Fomes officinialis Ames,Compared with the model group,the concentrations of serum lgG(IgG-S)and respiratory fluid lgG(IgG-R)in the administration group were relatively reduced(P<0.05),the expression of trypsin protease-activated receptor-2(PAR-2)was relatively weakened(P<0.05);the spleen coefficient and thymus coefficient were increased(P<0.05).Analysis of the Pearson correlation coefficients of the four indicators in the five groups,no clear correlation was found.After NMR and metabolomics analysis,compared with the model group,the VLDL,citrulline,lipid,glutamic acid,acetylglycoprotein,proline,arginine,Choline,α-glucose,glycerol,and unsaturated fatty acids increased;while lactic acid and alanine showed different trends at differentchemical shifts in serum of rats.Compared with the model group,valine,2,3-dihydroxyisoglutarate,dodecanoate,3-aminoisovalerate,proline,2-oxoisohexanoic acid,hypotaurine,mannitol,2-methylglycine,aspartic acid,hippuric acid,malic acid,and tyrosine increased;taurine and acetoacetic acid decreased in urine of rats.Conclusion:Fomes officinialis Ames can change the metabolism,restore immune function,enhance the expression of trypsin and PAR-2,and reduce the coefficient of spleen and thymus of rats with cold-damp syndrome.Cold and wet environments can cause significant changes in various metabolites in serum and urine,and affect multiple metabolic pathways.
