| Objective:Using data analysis technology to explore the core prescriptions for clinical prevention and treatment of atrial fibrillation and experimentally observe its effects on atrial fibrillation induction time,duration,serum cTnI,BNP,CRP content,atrial fibrosis and other indicators in paroxysmal atrial fibrillation rats Its possible efficacy mechanism.Methods: 1.Data mining part: Collect Chinese medicine prescriptions for prevention and treatment of atrial fibrillation in the Cardiology Department of the First Affiliated Hospital of Anhui University of Traditional Chinese Medicine from June 2016 to June 2019.After standardized Processing,they are converted into data that can be identified by calculation.Use Microsoft Office Excel 2016 and SPSS Statistic 24.0 performed association analysis and cluster analysis on the data,and finally obtained the medication laws and core prescriptions.2.Experimental part:(1)Rats were randomly divided into prevention group western medicine group,traditional Chinese medicine group,model group and normal group with the same sample size(10 rats);(2)The tail vein was injected with acetylcholine-calcium chloride solution(Ach 99μg / m L + Cacl2 10mg/m L)0.1m L/100g/d for 7 consecutive days,and the drug was administered according to the prevention group and the traditional Chinese medicine group 24.23g/Kg/d;Western medicine histiodone 54mg/Kg/d;model group was given the same amount of normal saline;the prevention group was given gastric administration before model making;the normal group did not intervene;ensure that the gastric administration and modeling time of each group were the same;(3)Using a biosignal acquisition system to trace the standard lead ECG before and after the rat experiment;and calculate the induction and duration of AF;(4)The serum cTnI,BNP,and CRP contents of each group of rats were determined according to the method of using the kit after the experiment;(5)Pathological changes of myocardial tissue cells were observed with masson staining under electron microscope;(6)Statistical analysis was performed using SPSS 24.0.Results: 1.Data amount mining part:(1)A total of 3358 medical cases were included,involving 201 flavors of drugs and 50370 medications,with an average of 15 flavors per case;(2)The top 15 drugs are Chenpi,Baizhu,Dangshen,Danshen,Shashen,Zhuru,Suanzaoren,Gansong,Yuanzhi,Pugongying,Yiyiren,Banxia,Fuling,Renshen,Gualoupi,the main effect is to Tiao Pi Hu Xin,An Shen Ding Ji,Xing Qi Hua Tan;(3)ent;the drugs are mostly flat and warm;the meridians are mainly spleen and stomach;(4)10 related medications as “Baizhu → Chenpi,Chenpi → Dangshen";(5)Two clustering results were obtained by cluster analysis,namely: "Sha Shen,Gansong,Danshen,Renshen,Chenpi,Baizhu,Zhuru,Pugongying,Dangshen,Yiyiren,Banxia,Suanzaoren,Yuanzhi,Shenqu”,“Maidong,Dihuang,Zhimu,Chuanniuxi,Longgu,Muli,Fuxiaomai,Fuling,Gualoupi,Tianma,Hehuanpi,Shanyao,Baishao,Gouqizi,Juemingzi,Jineijin”;2.Experimental part:(1)Atrial fibrillation induction time: Compared with the model group,the atrial fibrillation induction time in the prevention group,the Chinese medicine group,and the western medicine group was prolonged,and there was a difference(P<0.05).Compared with the Chinese medicine group,the prevention group could significantly extend the atrial There was no significant difference in the induction time of fibrillation(P<0.05);there was no statistical difference in the induction time of atrial fibrillation between the prevention group and the western medicine group(P>0.05);(2)Atrial fibrillation duration: The prevention group,the Chinese medicine group and the western medicine group significantly shortened the duration of atrial fibrillation and were different from the model group(P<0.05);the Chinese medicine group,the western medicine group and the prevention group had differences(P<0.05),the average value of the prevention group(4.14s)<the average value of the Chinese medicine group(5.85s);(3)Serum BNP,cTnI,and CRP levels: Compared with the model group,the serum BNP and cTnI levels in the other groups were lower and there were differences(P<0.05).The serum cTnI levels in the prevention group,western medicine group,and Chinese medicine group were not different from the normal group.Statistical significance(P>0.05);Compared with the prevention group,the Chinese medicine group was equivalent to the prevention group in reducing serum cTnI content(P>0.05).The Chinese medicine group was not as effective as the prevention group in reducing serum BNP content(P>0.05);Compared with the model group,the serum CRP content of each group was low and there was a difference(P<0.05);there was no difference in serum CRP content between the Chinese medicine group and the prevention group(P>0.05),but the average value of the prevention group(994.11pg/m L)<the average value of the traditional Chinese medicine group(1818.39pg/m L);there is a difference in CRP content between the traditional Chinese medicine group and the western medicine group(P<0.05);(4)Changes in myocardial pathology sections:Myocardial cells in the normal group are neatly arranged,oval in shape,and a few normal dotted blue areas in the outer matrix are normal collagen structures.Cells in the model group have disordered arrangement,enlarged gaps,strange shapes,dense blue and areas,and deep staining.The cells were distributed in a patch-like manner with the highest degree of myocardial fibrosis;the cells in the prevention group were partially deformed and arranged neatly.In addition to the normal cell results,the blue area was more than the normal group.Less,pathological changes under the microsc opeare better than the traditional Chinese medicine group,indicating that the prevention group can better prevent myocardial fibrosis;the western medicine group has most of the normal arrangement of cells,the shape can be,the overall orange-red,blue areas are dotted,that is,normal tissue The number of fibrosis is lower than that of the model group;a few of the flaky areas of the Chinese medicine group are disordered,the tissues are flaky necrosis,the color is uneven,the myocardial interstitial edema,the overall visual field is dark red,and the red dots are more numerous,blue The areas were scattered in stripes,indicating that there were more collagen fibers and muscle fibers than the normal group,but less than the model group.Conclusion: 1.Data mining part: The prescription reflects the rule of "spleen and spleen Tongzhi" medication,and two core prescriptions are summarized through data mining.Among them,"ShaShen,Gansong,Danshen,Renshen,Chenpi,Baizhu,Zhuru,Pugongying,Dangshen,Yiyiren,Banxia,Suanzaoren,Yuanzhi,Shenqu” are more in line with clinical prevention and treatment thinking;Named "Shenzhu Ningxin Decoctione";2.Experimental part:(1)Shenzhu Ningxin Decoction can be administered to the stomach before and after modeling,and the outcome indicators are different,which can play a preventive role;(2)Shenzhu Ningxin Decoctione prolongs the induction time of atrial fibrillation in rats,and has the effect of preventing the disease;(3)Shenzhu Ningxin Decoctione shortens the duration of atrial fibrillation and promotes the recovery of the disease;(4)Shenzhu Ningxin Decoction reduces serum CRP content in myocardial atrial fibrillation rats and reduces myocardial inflammation(5)Shenzhu Ningxin Decoctione inhibits atrial myocardial fibrosis and improves atrial structural remodeling;(6)There were no obvious adverse reactions in clinical application and experimental research of Shenzhu Ningxin Decoction. |
