BackgroundWith the change of people’s life style,patients with hypertension have been increasing day by day.Hypertension is an important risk factor for cardiovascular and cerebrovascular diseases and kidney diseases.Its complications,such as coronary artery disease,kidney failure,stroke,have a high mortality and disability rate.Renal hypertension is a kind of hypertension,which belongs to secondary hypertension.And renal hypertension can be divided into renal vascular hypertension and renal parenchymal hypertension,both of which have varying degrees of renal artery changes.Auricularia auricula,as a common edible fungus,has delicious taste,high nutritional value and high medicinal value.It is recognized worldwide as health food.Auricularia auricula polysaccharide(AAP)is the main component of Auricularia auricula.It can extract AAP from a certain way and study its effect on renal hypertensive rats.ObjectivesThe effects of Auricularia auricula polysaccharide(AAP)on blood pressure,kidney and heart in renovascular hypertensive rats were studied by animal experiment,noninvasive tail blood pressure measuring instrument,automatic chemiluminescence analyzer and histopathology.Methods1.Auricularia auricula polysaccharide(AAP)was prepared by high temperature steam extraction.2.The two kidney one clip renovascular hypertension(RHR)rat model has the advantages of simple modeling and high success rate.It is the most widely used in hypertensive rat models.Therefore,RHR rat models were used in this study,and the modelgroup,sham operation group,Kato Pury group,AAP high dose group and AAP low dose group were established respectively.3.The standard II lead ECG of each group was compared and the difference was observed.4.The renin-ang-aldosterone system(RAAS system)was activated,which led to the increase of blood pressure in experimental rats.Through the determination of Renin,AII and ALD in plasma,the data collected were compared by using statistical method,reflecting the change degree of renal hypertension rats.5.The model group,the fake test group,the Kato Pury group,the black fungus polysaccharide high and low dose group,each group of rats kidney tissue and heart tissue,weighing,sectioning,staining,observed and compared the difference between the groups.Results1.Measurement of blood pressure of rats in each group.The experimental results show that compared with the model group,the blood pressure of captopril group,AAP high-dose group and AAP low-dose group decreased significantly(P<0.01 or P<0.05).Compared with captopril group,The blood pressure of high dose AAP group decreased significantly(P<0.05),but the difference of low dose AAP group was not significant.The plasma Renin of captopril group and AAP high and low dose group was lower than that of model group,the difference was statistically significant(P<0.01 or P<0.05).The AII of plasma in captopril group and AAP high and low dose group was lower than that in model group,the difference was statistically significant(P<0.01 or P<0.05).Aldosterone content in plasma of model group was significantly higher than that of Captopril Group and AAP group,the differences were statistically significant(P<0.01 or P<0.05).2.Changes of electrocardiogram in renovascular hypertensive rats.The ECG of the rats in each group was detected by standard II lead.Electrocardiogram.The S-T segment elevation,QRS wave width and T wave were increased in themodel group.3.Histopathological changes of kidney and heart in rats of each group.The kidney and heart tissues of rats in each group were stained with HE,and the results were compared.The results of HE staining showed that the structure of glomeruli and renal tubules in the Sham-operation Group and the Blank Control Group was clear and there was no abnormal pathological change,while some glomeruli in the model group were atrophic with fibroproliferation,and the above symptoms were relieved in the captopril group and AAP high and low dose group.In the Sham Operation Group and Blank Control Group,the arrangement of myocardial cells was uniform,the thickness of myocardial fibers was roughly the same,and the organization was clear;in the model group,the arrangement of myocardial cells was disordered,with edema,thickening,enlargement and rupture;in the captopril group and AAP high and low dose group,the above symptoms were relieved.Conclusions1.AAP can significantly lower Renin,Angiotensinogen and aldosterone levels which relative plasma index of hypertension.2.AAP can significantly improve the renal and cardiac injury in RHR rats,and is beneficial to the recovery of cardiac function.3.AAP can significantly reduce the blood pressure of RHR rats,and high doses of it were better than Captopril’s.
