Objectives:To explore the effect of dagreen on body weight,blood lipids,blood glucose and insulin and its relationship with adipose factor chemerin by observing the effects of dapagliflozin on glucose and lipid metabolism,serum chemerin level and liver tissue chemerin expression in type 2 diabetic rats.Methods:20 healthy male SD rats were randomly divided into normal group(NC group)and experimental group.Rats in normal group were fed with normal diet,while rats in experimental group were fed with high-fat diet,and all rats were free to eat and drink.Four weeks later,the rats in the NC group were intraperitoneally injected with sodium citrate solution(10ml/kg)of 0.1mol/L,and the rats in the experimental group were intraperitoneally injected with STZ(35 mg/kg,i.p.).The diabetic rat model was induced and established.7 days later,the diabetic rat model was considered successful if the FPG,FPG was higher than 11.1mmol/L,accompanied by polydipsia,polyuria and polyuria.12 rats were successfully modeled,with a success rate of 75%.Twelve successful rats were randomly divided into two groups: T2 DM model group(T2DM group)and dapagliflozin group(DAPA group).The rats in NC group and T2 DM group were treated with 2ml saline,while those in DAPA group were treated with dapagliflozin 1mg / kg,dissolved in 2ml saline and given intragastric administration once a day.During the experiment,all rats continued to be fed with normal diet for 6 weeks.All rats were weighed every other week.Before and after intervention,serum chemerin and fasting insulin were measured by ELISA method,fasting blood glucose was measured by glucose oxidase method,blood lipids were measured by spectrophotometry,direct method and enzyme method,and HOMA-IR and HOMA-β were calculated.After intervention,the expression of chemerin in liver was detected by WB method.The experimental data were analyzed by t-test,ANOVA,LSD and pearson correlation analysis.Results:After successful modeling and before intervention,compared with NC group,the BW,FPG,FINS,HOMA-IR,TG,TC,LDL and serum chemerin levels in T2 DM group and DAPA group increased,while high density lipoprotein and HOMA-β decreased,but there was no significant difference between T2 DM group and DAPA group.Compared with T2 DM group,BW,FPG,FINS,HOMA-IR,TG,TG,serum chemerin level decreased,high density lipoprotein and HOMA-β increased,liver chemerin protein expression decreased in DAPA group after intervention,among which TG decreased significantly,LDL slightly increased,but there was no statistical difference.Chemerin protein expression in liver tissue decreased in DAPA group.The correlation analysis between chemerin and various indexes showed that before and after intervention,it was positively correlated with serum chemerin and BW,TG,TC,LDL,FPG,FINS and HOMA-IR,and negatively correlated with HDL and HOMA-β,and the content of chemerin in serum was positively correlated with the expression of this protein in liver tissue.Conclusion:The level of chemerin in serum and liver tissue of type 2 diabetic rats is higher than that of non-diabetic rats.As a new hypoglycemic drug,dapagliflozin can improve the disorder of glucose and lipid metabolism in type 2 diabetic rats,which may be related to down-regulating the level of chemerin protein in serum and liver tissue.
