Objective:The quality comparison study of Mongolian medicine Scabiosa comosa Fisch(including Mongolia)from different producing areas was carried out to provide scientific basis for the formulation of quality standard of Mongolian medicine and to ensure the clinical efficacy and the safety of compound preparation applicationMet hods:The microscopic identification method was used for the qualitative identification of Mongolian medicine Scabiosa comosa Fisch,the quantitative determination of Chlorogenic acid,Caffeic acid,Luteolin,Sweroside,Isoquercitrin,Galuteolin,Isochlorogenic acid A,Apigenin-7-O-Glucoside,Apigenin and other active components was studied by HPLC.Chromatographic column Welchrom-C18(4.6mm×300,5?m),mobile phase acetonitrile-0.3%phosphoric acid water,detection wavelength of 254nm,flow rate of 1mL/min,column temperature of 40?,injection volume of 10?L.The fingerprints of 24 batches of Scabiosa comosa Fisch from different producing areas were studied by HPLC.The chromatographic conditions were as following:Column:Welchrom-C18(4.6mm×300,5?m),mobile phase:acetonitrile(B)-0.3%phosphoric acid water(A)gradient elution(B:0.01?25min,5%?8%;25?50min,8%?15%;50?75min,15%?19%;75?100min,19%?24%;100-110min,24%?7%;110-115min,7%-4%;115-120min,4%?0%)According to the determination method of water content,ash content,acid insoluble ash content and water soluble extract in Chinese pharmacopoeia,routine examination was carried outResults:The fibrous characteristics of glandular hair,non-glandular hair,pollen grains,style epidermal cells,petal epidermal cells,calcium oxalate crude crystals in the powder of Scabiosa comosa Fisch were identified by microscope,The contents of Chlorogenic acid,Caffeic acid,Luteolin,Sweroside,Isoquercitrin,Galuteolin,Isochlorogenic acid A,Apigenin-7-O-Glucoside,Apigenin in Scabiosa comosa Fisch from different habitats were determined simultaneously by HPLC,Results Chlorogenic acid,Caffeic acid,Luteolin,Sweroside,Isoquercitrin,Galuteolin,Isochlorogenic acid A,Apigenin-7-O-Glucoside,Apigenin were found respectively in 2576?15456 ng,27.35?164.1 ng,171.2?1027.2 ng,ng 106?636,148?888 ng,ng 819?4914,840?5040 ng,226?1356 ng,86?516 ng had good linear relationship with the respective peak area(r value was 0.9999?0.9994?0.9991?0.999?0.9994?0.9998?0.9992?0.9998?0.9997).The accuracy,stability,and repeatability of the RSD%were all less than 2.0%,and the recovery rates were 96.4%?95.43%?96.17%?96.22%?95.54%?96.42%?96.62%?95.57%?95.80%,and RSD%was 0.24%?0.72%?0.46%?0.52%?0.62%?0.82%?0.59%?0.53%?0.54%(n=6)The HPLC characteristic chromatogram of Mongolian medicine Scabiosa comosa Fisch was established,with S12 as the reference.The results showed that there were 22 common peaks,and the similarity analysis was carried out by using the software of similarity evaluation system of traditional Chinese medicine chromatogram fingerprint(version A).The similarity of 24 batches of Scabiosa comosa Fisch was?0.900.The results of all inspection items meet the requirementsConclusion:Several identification characteristics of Scabiosa comosa Fisch were identified by microscopic identification method,which provided reference for the establishment of quality standard of Mongolian medicine Scabiosa comosa Fisch.HPLC method was used to establish the HPLC fingerprint method for the determination of Chlorogenic acid,Caffeic acid,Luteolin,Sweroside,Isoquercitrin,Galuteolin,Isochlorogenic acid A,Apigenin-7-O-Glucoside,Apigenin and other nine components of Mongolian medicine lannifera.This method is simple to operate,good repeatability,and has both qualitative and quantitative functions.According to the results,it can provide reference for the establishment of the quality standard of Mongolian medicine Scabiosa comosa Fisch,and also lay a foundation for the resource development and utilization of this medicinal material.
