ObjectivesThe aim of this study was to investigate the expression of triplet motif 14(TRIM 14)in benign thyroid nodules and malignant thyroid nodules,The relationship between the two attempts to help improve the diagnosis of benign thyroid nodules and thyroid malignant nodules;To analyze the expression of TRIM 14 in papillary thyroid carcinoma and its clinicopathological features.it also provides a theoretical basis for the choice of treatment options for patients with thyroid papillary carcinoma.Methods1.Clinical data collection:A total of 66 patients(17 benign thyroid nodules,49 thyroid malignant nodules)were selected from Yunnan tumor Hospital from January 1,2010 to December 31,2016.Case system collects data on patients with thyroid papillary carcinoma(clinical characteristics include:age,gender,AJCC tumor stage(8th edition)when the patient first visited our hospital,and whether iodine-refractory thyroid cancer was evaluated after 131I treatment;The pathological features include:the largest diameter of the lesion during surgical resection,whether the capsule is infiltrated with the naked eye,whether there is lymph node metastasis,whether there is distant metastasis,multifocal;preoperative thyroid-related hormone levels include:T3,T4,TSH,Tg,TgAb,TMAb,FT3,FT4,rT3.)2.Immunohistochemical staining:the corresponding paraffin-embedded tissue specimens of 66 thyroid nodules patients were selected,and the expression of TRIM14 protein was semi-quantitatively determined by immunohistochemistry.3.Statistical analysis:statistical software SPSS20.0 for analysis;non-parametric rank sum test(Wil Coxon rank sum test)using two independent samples to analyze TRIM 14 in benign thyroid nodules and thyroid malignant nodules The difference in expression between patients;clinical,pathological features and serum thyroid hormone levels were converted into two-category data according to the relevant reference indicators,and then the non-purametric rank sum test(Wil Coxon rank sum test)for the comparison of two independent samples.P<0.05 Indicates that the difference is statistically significant.Results1.TRIM14 protein expression:TRIM14 protein staining positive in thyroid tissue was brown,brownish yellow,light yellow,localized in the cytoplasm;2.In 17 cases of benign thyroid nodules,TRIM 14 expression was negative in 14 cases(82.35%),weakly positive in 3 cases(17.65%),medium positive in 0 case(0%),strong positive in 0 case(0%);49 cases of thyroid In malignant nodules,TRIM14 was negative in 15 cases(30.61%),weakly positive in 21 cases(42.86%),medium positive in 11 cases(22.45%),and strong positive in 2 cases(4.1%).After statistical analysis,the expression of TRIM 14 in benign thyroid nodules and malignant nodules was statistically significant(z=-3.71,P<0.001),and the intensity of TRIM 14 expression in malignant thyroid nodules was higher than that of benign thyroid nodules(The mean rank of malignant thyroid nodules was 38.30,and the mean rank of benign thyroid nodules was 19.68);3.The relationship between the expression of TRIM14 and the clinical features of patients with papillary thyroid carcinoma:the expression of TRIM 14 in papillary thyroid carcinoma is related to whether patients have iodine refractory(z=-2.33,P=0.02),and non-iodine The expression of TRIM14 was higher in the refractory thyroid cancer group(rank mean value of iodine-refractory differentiated thyroid cancer group 16.45,mean rank of non-iodine-refractory thyroid cancer group 24.55);and sex with patients(z=-0.26,P=0.79),age(z=-0.74,P=0.46),and AJCC tumor stage(z=-0.89,P=0.37)were not related.4.The relationship between the expression of TRIM 14 and the pathological features of papillary thyroid carcinoma:the expression of TRIM 14 in the papillary thyroid carcinoma and the maximum diameter of the lesion(z=-1.58,P=0.12),whether there is capsule infiltration(z=-1.48,P=-0.14),lymph node metastasis(z=-0.52,P=0.60),distant metastasis(z=-1.56,P=0.12),multifocal(z=-0.50,P=0.62).5.The relationship between TRIM 14 expression and serum thyroid hormone levels in patients with papillary thyroid carcinoma:TRIM14 expression in thyroid papillary carcinoma and preoperative serum thyroid hormone levels(T3(z=-0.65,P=0.51))),T4(z=-0.11,P=0.91),TSH(z=-1.11,P=0.27),Tg(z=-0.68,P=0.50),TgAb(z=-0.31,P=0.76),TMAb(z=-0.53,P=0.60),FT3(z=-0.17,P=0.87),FT4(z=-1.44,P=0.15),rT3(z=-0.87,P=0.38)).Conclusion(s)1.TRIM 14 protein expression in thyroid malignant nodules is significantly higher than benign thyroid nodules,suggesting that TRIM 14 can be used for the differential diagnosis of benign and malignant thyroid nodules,but TRIM 14 is not a specific marker.2.If the high expression of TRIM 14 protein in the cancer tissue of patients with thyroid papillary carcinoma often indicates that the patient’s postoperative 131I treatment is often better.3.TRIM 14 protein expression and thyroid papillary carcinoma patients gender,age,tumor stage(AJCC 8th Edition),tumor maximum diameter,multifocal,lymph node metastasis,distant metastasis,combined with capsule infiltration,serum thyroid hormones(T3,T4,TSH,Tg,TgAb,TMAb,FT3,FT4,rT3)are irrelevant.
