| Orychophragmus violaceus(L.)is a herb of the cruciferous genus.It is used for the treatment of children with jaundice,women’s chyle,and common heat toxic diseases.Modern research has found that O.violaceus has good activity in anti-oxidation and anti-inflammatory,and has certain liver-protecting effects.In this paper,the phenolic chemical constituents of O.violaceus were studied by modern chromatographic techniques and separation methods.At the same time,the content determination and fingerprinting of O.violaceus were studied for the first time.The main research contents and results are as follows:1.The extraction and separation of the aqueous extracts of O.violaceus seeds were carried out by column chromatography.The separation was determined by UV,IR,MS,1H NMR,13C NMR,HSQC,HMBC,1H-1H COSY,NOESY and other physicochemical properties.14 compounds were obtained:Orychophragmuspine A(1),Orychophragmuspine B(2),Orychophragmuspine C(3),Orychophragmuspine H(4),2-amino-5-hydroxybenzoic acid(5),3’,5’,5,7-tetrahydroxydihydroflavone(6),catechin(7),quercetin(8),salicylic acid(9),p-hydroxycinnamic acid(10),vanillic acid(11),original Catechin(12),caffeic acid(13),syringic acid(14),Epigoitrin(15).Among them,1-4 are new alkaloid compounds,and 5-15 are known compounds.2.In this experiment,TLC of different batches of O.violaceus seeds were tested under the same amount of points.The results showed that the spots in different places were different,indicating that the corresponding contents were different In addition,some studies have found that Epigoitrin has certain physiological activity and is high in O.violaceus,so it can be used as a test standard for the determination of O.violaceus.This experiment uses Waters ACQUITY ultra-high performance liquid.The chromatograph was used to analyze the seeds of O.violaceus,and the content of 15 batches of Epigoitrin was determined,and the range was 0.63%~0.92%.The results showed that the content of Epigoitrin in O.violaceus could be used as one of the quality control methods of O.violaceus medicine.From the perspective of quality control of Chinese herbal medicines,the systematic and comprehensive determination of O.violaceus seeds was qualitative and comprehensive.The analysis lays a scientific foundation for the subsequent Chinese medicine production process and the application of O.violaceus in the clinical practice of Chinese medicine.3.In this paper,UPLC(Ultra High Performance Liquid Chromatography)was used to detect the fingerprints of 15 batches of O.violaceus seeds from different producing areas.At the same time,the method of precision,stability and repeatability of the experimental methods was investigated.The most efffective conditions were used as conditions for this experiment.The results of various experiments show that the UPLC fingerprint method of O.violaceus is accurate and feasible,and can be used as a method to quickly analyze the chemical constituents of O.violaceus,and provide reference for the future research of medicinal plants in fingerprint.In summary,this thesis systematically studied the phenolic compounds of O.violaceus and established a quality control method for the phenolic compounds of O.violaceus.At the same time,fingerprint research on the medicinal materials with different harvesting time in different producing areas provides a scientific basis for the future research of O.violaceus. |
