Objective: To select the superior strains of Codonopsis pilosula and prepare the biotransformation products of Codonopsis pilosula and study its chemical constituents.To screen the purification method of total saponins from biotransformation products of Codonopsis pilosula and prepare the dropping pills of purified product.Methods: The biotransformation technology was used to select the superior strains of Codonopsis pilosula and to prepare the biotransformation products of Codonopsis pilosula.The biotransformation products were extracted with 70% ethanol.The extractive was separated and purified by silica gelcolumn chromatography,ODS column chromatography and semi-prepared liquid phase.The structures were elucidated by physical and chemical methods and spectral data.The purification methods of total saponins from biotransformation products of Codonopsis pilosula were screened by using D101 macroporous resin with static adsorption rate,sample concentration,diameter height ratio,dynamic adsorption rate and eluant concentration as inspection indexes.Response surface curve method was used to determine the preparation process of dropping pills.Results: Ganoderma lucidum was selected as the superior strains of Codonopsis pilosula,and the biotransformation products of Codonopsis pilosula were prepared.Eight compounds were isolated and identified from biotransformation products of Codonopsis pilosula,including:Vanilli(1),Pentadac-5-enoic acid methyl ester(2),Methyl(5z,9z)-(±)-2-hydroxy-5,9-eicosodienoate(3),Stigmasterol(4),?-sitosterol(5),25(R)-5?-spirostan-1?,3?-diol(6),ligustrin B(7),9,12-cyclomulin-13-ol(8).The purification process of total saponins from biotransformation products of Codonopsis pilosula was determined by D101 macroporous adsorption resin as follows: adsorption time 150 min,concentration of sample 0.5g/ml,ratio of diameter to height 1:20,amount of sample 1 BV,70% as eluent.Three factors,temperature of drug solution,ratio of drug to matrix and ratio of PEG4000 to PEG6000,were selected by single factor investigation.Response surface curve method was used to determine the forming process of dropping pills as follows: liquid temperature 80 ?,drug: matrix = 1:4,PEG4000: PEG6000 = 1:2.Conclusion: Provided new ideas for the development and utilization of traditional Chinese medicine through the preparation of biotransformation products of Codonopsis pilosula.The material composition of biotransformation products of Codonopsis pilosula has been further explored.Purification of total saponins from fermentation products of Codonopsis pilosula by D101 macroporous resin and preparation of dropping pills,laid a foundation for the innovation and development of new preparations of Codonopsis pilosula.
