BackgroundRadiation therapy is an important treatment method for esophageal cancer.The common reason for failure of radiotherapy is metastasis or local recurrence after radiotherapy,which suggests that different esophageal cancer patients have different sensitivity to radiation.procollagen-lysine,2-oxoglutarate5-dioxygenase 2(PLOD2)is a collagen-modifying enzyme that can promote the progression of esophageal cancer by changing the cross-linking pattern of collagen fibers in the extracellular matrix,but its relationship with the radiosensitivity of esophageal cancer has not been elucidated.ObjectiveToinvestigate the relationship between the expression level of PLOD2 in esophageal squamous cell carcinoma and its clinicopathological characteristics,radiosensitivity and prognosis,and to find molecular markers to predict the radiosensitivity of esophageal cancer.MethodsRetrospective analysis of clinical data of 90 patients with esophageal squamous cell carcinoma who underwent radical radiotherapy at the xx hospital from May 2015 to May 2019,The expression of PLOD2 in tumor tissues before radiotherapy was detected by immunohistochemistry,and its relationship with clinicopathological parameters,radiosensitivity and prognosis were analyzed.Results1.The level of PLOD2 expression in 90 esophageal squamous cell carcinoma tissues was significantly correlated with tumor clinical stage,lymph node metastasis,and distant metastasis(P <0.05),and had no correlation with patient gender,age,lesion location,lesion length,and T stage(P > 0.05).2.90 patients with esophageal squamous cell carcinoma underwent esophageal barium meal X-ray examination to evaluate the efficacy within 3 months after completion of radiotherapy.There were 51 cases in the radiosensitivity group(CR +PR),39 cases in the radiation resistance group(NR),and the PLOD2 expression level in the radiation resistance group Higher than the radiosensitive group,the difference was statistically significant(P <0.05).3.Kaplan-Meier univariate analysis showed that PLOD2 expression level was significantly correlated with disease-free survival in patients with esophageal squamous cell carcinoma(PFS)(P <0.05).Multivariate analysis of the Cox proportional hazards model showed that PLOD2 expression level showed that PLOD2 expression level Independent prognostic factors for esophageal squamous cell carcinoma(HR=3.395,95%CI=1.974~5.838,P <0.001).Conclusion1.The expression of PLOD2 has significant correlation with the clinicopathological characteristics of tumors.PLOD2 may promote the occurrence and development of esophageal squamous cell carcinoma.2.The expression level of PLOD2 in esophageal squamous cell carcinoma tissue is negatively correlated with the radiosensitivity of esophageal squamous cell carcinoma.This indicates that PLOD2 may be a molecular marker for measuring radiosensitivity of esophageal squamous cell carcinoma.3.Patients with high expression of PLOD2 after esophageal squamous cell carcinoma have poor prognosis after radiation therapy,suggesting that PLOD2 may be a predictor of prognosis of esophageal squamous cell carcinoma with radiotherapy.
