| Inflammation is the basic disease of many major diseases in human beings,and it is of great significance to control the development of inflammation.Neutrophils are an important part of innate immunity and play an important role in regulating inflammation and immune response.Neutrophils are important inflammatory cells that cause human inflammatory diseases.When stimulated by inflammation,neutrophils leave the blood and are activated by many stimulants,such as cytokines,growth factors,and pathogen products that may extend the life of neutrophils.Neutrophils buoyed by the pathogen,causing the neutrophils life extension,which could help to adaptive immunity inflammation subsided or transition,but will intensify neutrophils to inflammation parts remo ved and then increase the phagocytosis of neutrophils,result in tissue damage to cells "innocent" thus aggravating inflammation.Biological activity of hyaluronic acid(Bioactive hyaluronic acid,B-HA)by repeated D-glucuronic acid and N-acetyl-D-glucosamine disaccharides units repeatedly can be connected with the alternation of sulfated amino sugar,have be en found and confirmed that B-HA is bacterial endotoxin antagonist,by combining with human toll-like receptors and receptor CD44,reduce inflammation factor secretion,to treat inflammation,but its mechanism is unclear.One of the main active ingredients of Astragalus Membranaceus is polysaccharide,and berberine is one of the main bioactive ingredients of Rhizoma coptidis.Studies have shown that thes e two drugs have good effects on tumor treatment,controlling inflammation,regulating immunity and fighting infection,etc.However,the anti-inflammatory mechanism of these two drugs is still not very clear.In this study,lipopolysaccharide(LPS)was used to stimulate human neutrophils to determine its effect on the phagocytosis and removal of human neutrophils and to stimulate the production of inflammation.At the same time,choose B-HA,Radix Astragali,Rhizoma Coptidis,three kinds of medication by flow cytometry instrument test groups of samples processed neutrophil phagocytosis of fluorescent microspheres,analysis of different drugs for neutrophils phagocyte phagocytosis rate difference,developed the influence of different drugs for human neutrophil phagocytosis;Using agarose droplet method,preparation of neutrophils agarose mixture,forming gum drops,selects the biological activity of hyaluronic acid(B-HA),radix astragali,rhizoma coptidis three drugs to deal with add to gum drops,judging by inverted microscope gum drops of neutrophils in the change of the migration distance,the gum drops can be determined through Image J software neutrophils removed outside the area,developed different drugs for people neutrophils to remove the effects of exercise.The main findings are as follows:(1)Study on the effects of B-HA,Astragalus Membranaceus and Rhizoma Coptis alone or in combination on the phagocytosis of human neutrophilsLPS can significantly promote the phagocytosis of human neutrophils,thus stimulating the production of inflammation.B-ha slightly promoted the phagocytosis of human neutrophils.Coptis chinensis had a slight inhibitory effect on the phagocytosis of human neutrophils.The combination of B-HA+coptis can slightly promote the phagocytosis of human neutrophils.Astragalus group,BHA+astragalus group,and B-HA+astragalus+rhizoma coptis group all promoted the phagocytosis of human neutrophils,and the combined use of three drugs,B-HA,astragalus and rhizoma coptis,had a strong promoting effect on the phagocytosis of human neutrophils.(2)Effects of B-HA,Astragalus Membranaceus and Rhizoma Coptis alone or in combination on human neutrophil migrationWhen the serum content inside and outside of the droplet is the same,LPS can significantly promote the removal of human neutrophils,and cause the neutrophils to move and accumulate outside the droplet,thus stimulating the production of inflammation.B-HA can promote the movement of human neutrophils.Coptis chinensis significantly inhibited the removal of human neutrophils.The combined treatment of B-HA+coptis can slightly inhibit the removal of human neutrophils.Astragalus group,B-HA+astragalus group,and B-HA+astragalus+rhizoma corydalis group all promoted the movement of human neutrophils,and the combined use of three drugs,BHA,astragalus and rhizoma corydalis,had a strong promoting effect on the movement of human neutrophils.When the serum content outside the droplet is higher than that inside the droplet,LPS can significantly promote the removal of human neutrophils,and cause the neutrophils to move and accumulate outside the droplet,thus stimulating the production of inflammation.B-HA can promote the movement of human neutrophils.Coptis chinensis significantly inhibited the removal of human neutrophils.The combined treatment of B-HA+ coptis can slightly inhibit the removal of human neutrophils.Astragalus group,B-HA+astragalus group,and B-HA+astragalus+rhizoma corydalis group all promoted the movement of human neutrophils,and the combined use of three drugs,BHA,astragalus and rhizoma corydalis,had a strong promoting effect on the movement of human neutrophils. |
