Objective:To explore the expression of glucose transporter protein 1(GLUT1)in placental of severe preeclampsia in hypertensive disorders of pregnancy and its relationship with maternal lipids in the third trimester of pregnancy.Methods:Totally 31 pregnant women admitted to obstetrics department of affiliated hospital of North Sichuan Medical College from July 2018 to November 2019 were recruited.Among them,15 patients with severe preeclampsia were taken as experimental group and 16 healthy pregnant women as control group.The placental tissue was collected within 10 minutes after delivery of the placenta during cesarean section,using a sterile tissue scissors to cut the placental tissue in the direction of the umbilical cord and the placental edge at about 1cm*1cm*1cm,detecting its GLUT1 expression and calculating the average optical.At the same time,3ml of venous blood was collected from fasting state before cesarean section in two groups of pregnant women,detecting the triglyceride(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL),low density lipoprotein cholesterol(LDL)and exploring their relationship with GLUT1 expression.Result:1.GLUT1 is expressed in placenta tissues of severe preeclampsia and healthy pregnancy.Glut1 is mainly concentrated on syncytiotrophoblast cell membrane of placenta.The cell membrane of severe preeclampsia is deeply stained,while that of healthy pregnancy is slightly lighter.2.The average optical of GLUT1 in the center and edge of the placenta of the severe preeclampsia group were 0.0103 ±0.003 and 0.0082±0.002 respectively;the average optical of the center and edge of the placenta in the healthy pregnancy group were 0.0075±0.002 and 0.0063±0.002 respectively.There is no significant difference.(P>0.05).3.The average optical of GLUT1 in the central placenta of severe preeclampsia group and healthy pregnancy group were 0.0103±0.003 and 0.0075±0.002 respectively,The difference is significant(P<0.05);the average optical of GLUT1 in placental margin of severe preeclampsia group and healthy pregnancy group were 0.0082±0.002 and 0.0063± 0.002 respectively,The difference is significant(P<0.05).4.Comparison of blood lipids before cesarean section in severe preeclampsia group and healthy pregnancy group,TG were 4.60±1.7mmol/L and 4.41±1.6mmol/L,TC were 7.36± 2.4mmol/L and 6.94±1.1mmol/L,HDL were 1.87±0.5mmol/L and 1.83± 0.4mmol/L,and LDL were 4.07±1.7mmol/L and 3.44±1.2mmol/L,respectively.There is no significant difference between the indexes(P>0.05).5.There is no significant correlation between the expression of GLUT1 in the central placenta of severe preeclampsia and TG,TC,HDL,and LDL in blood lipids in the third trimester of pregnancy(r were 0.178,0.328,0.079 and 0.275 respectively,P>0.05).There is no significant correlation between the expression of GLUT1 in the marginal placenta of severe preeclampsia and TG,TC,HDL and LDL in maternal blood lipids in the third trimester of pregnancy(r were-0.229,-0.113,-0.189 and-0.047 respectively,P>0.05).Conclusion:1.GLUT1 is expressed in severe preeclampsia and healthy placental tissues,and is mainly concentrated on the placental syncytiotrophoblast cell membrane.2.GLUT1 expression in placenta of severe preeclampsia increased.This increase may be related to placental ischemia,degree of hypoxia and oxidative stress reaction in severe preeclampsia.3.Serum lipids in pregnant women with severe preeclampsia did not show significant high-fat state in the third trimester of pregnancy,and there is no significant correlation between the expression of placental GLUT1 and blood lipids in the third trimester of pregnancy.
