Expression Of SIRT2 And NLRP3 In The Blood Vessels Of Atherosclerosis Patients And Their Effects On Inflammation

Objective:To observe whether SIRT2 can be expressed in human aorta,and to study the expression of SIRT2 in aorta with different degrees of atherosclerosis.Methods:1.Organization:the extraction of the extraction of aortic dissection group according to the severity of atherosclerosis is divided into the experimental group 1,group 2 and normal aorta tissue(aortic dissection of traumatic factors)in a liquid nitrogen or-80? refrigerator,and tag information.2.Aortic dissection edge blood vessels(atherosclerosis extent relatively light)marked as experimental group 1,aortic intimal tear department blood vessels(atherosclerosis degree relatively serious)marked as experimental group 2,normal aorta vascular tissue(aortic dissection of traumatic factors)as control group,respectively,to be divided into 60 mg group,partial shipments and save.3.Western blotting was used to detect the protein expressions of SIRT2 and NLRP3 in the vessels at the edge of aortic dissection(relatively mild degree of atherosclerosis),the vessels in the aortic intimal tear(relatively severe degree of atherosclerosis),and the normal aortic vessels.4.Real-time PCR was used to detect the mRNA expressions of SIRT2 and NLRP3 in the vessels at the edge of aortic dissection(relatively mild degree of atherosclerosis),the vessels in the aortic intimal tear(relatively severe degree of atherosclerosis)and normal aortic vessels.Results:1.SIRT2 and NLRP3 are expressed in aortic tissue in atherosclerosis.2.In the real-time PCR experiment,SIRT2 and NLRP3 gene expression levels in aortic tissue of atherosclerosis showed an increasing trend compared with normal tissue,and the expression level gradually increased with the deepening of atherosclerosis.3.Western boltting experiment showed that,compared with normal tissues,the expression levels of SIRT2 and NLRP3 proteins in aortic tissue of atherosclerosis showed an increasing trend,and the expression level gradually increased with the deepening of atherosclerosis.Conclusion:Compared with normal aortic tissue,the expressions of SIRT2 and NLRP3 in aortic tissue of atherosclerosis showed an increasing trend,and with the aggravation of atherosclerosis,the expressions of SIRT2 and NLRP3 in aortic tissue gradually increased.
