Objective(s):This experiment uses flow cytometry to detect the cytokines’ expression levels of in patients with acute myeloid leukemia(AML)during initial diagnosis,infection,and complete remission,those cytokines iclude IL-1β,IL-2,IL-4,IL-5,IL-6,IL-8,IL-1β,IL-10,IL-12p70,IFN-γ,IFN-α,and TNG-α,to explore the significance of cytokine level changes in various disease states of AML.Methods:Selecting the period from June 2019 to January 2020,88 AML patients diagnosed and treated in Yunda Hospital(the first affiliated hospital of Kunming Medical University)were collected as the experimental group,of which 21 patients with newly diagnosed AML were newly diagnosed uninfected group,and 20 patients with newly diagnosed AML with infection were newly diagnosed group Eight CR AML patients with concurrent infection were CR infection group,39 CR AML patients were CR uninfected group,and the control group was 19 concurrent health checkups.3 ml of peripheral blood were collected from each of the five groups(experimental groups and the control group),and 12 items of cytokines were detected by flow cytometry,and the expression levels were recorded.SPSS 17.0 software was used to analyze relevant experimental data.When P<0.05,the difference was statistically significant.Results:1.Compare to the control group,the levels of IL-1β,IL-8 and IL-6 in the newly diagnosed AML uninfected group were significantly higher(P<0.05).and IL-2,IL-10,IL-17,IL-4,IL-5,IFN-y,TNG-α,IL-4,IL-12P70 were also slightly higher(P<0.05).2.The lower level of IL-8 in the AML undiagnosed group was related to the lower white blood cell count at the same time(r=-0.57;P<0.05).3.In the CR uninfected group of AML patients,the level of those cytokines iclude IL-1β,TNG-α,IL-5,IFN-y,and IL-4 were slightly higher than those in the control group(P<0.05),and the overall cytokine levels were close to the control group.4.Compare to non-infected group,the levels of IL-8 and IL-6 in the patients with AML at the initial diagnosis were significantly higher(P<0.05),and the levels of IL-1β,IL-2,IL-10,and IL-17 were also slightly higher than CR uninfected group(P<0.05).5.The level of IL-8 in the newly diagnosed AML infection group was significantly higher than that in the newly diagnosed uninfected group(P<0.05).6.The levels of IL-8 and IL-6 in the AML-CR infection group were significantly higher than those in the CR uninfected group(P<0.05),and the levels of IL-1β,IL-2,IL-10 and IL-12p70 were slightly higher than those in the CR uninfected Group(P<0.05).7.Higher body temperature in the newly diagnosed AML infection group was associated with higher IL-8 levels(r=0.57;P<0.05).8.The higher body temperature in the CR infection group was associated with higher levels of IL-6(r=0.81;P<0.05)and IL-10(r=0.83;P<0.05).Conclusion(s):In the initial diagnosis,infection,and CR of AML,the multi-cytokine levels we detected were partially abnormal,and we speculated that this abnormality may be related to the disease state.In the occurrence and development of AML,IL-1β,IL-6,IL-8 may be an important component of cytokine disorders.AML disease states suppress the body’s immune response,and in AML patients,IL-8 levels seem to be more closely related to infection.Cytokine detection may be an effective method to assist in the diagnosis and evaluation of AML or its infection,but further research is still needed.
