Objective:1.To obtain experimental evidences that Hematological malignancy serum-derived exosomes stimulate drug resistance and migration of liver cancer cells and cervical cancer cells.Methods:1.Total Exosome Isolation Reagent was used to extract human serum-derived exosomes,include healthy people and people with hematological malignancy.2.The morphology of the exosomes was observed by transmission electron microscopy(TEM).3.Western blot was used to detect the marker protein of human serum-derived exosomes.4.Western blot was used to detect the content of P-gp and MMP-2 protein in human serum-derived exosomes.5.Western blot was used to detect the content of P-gp protein in Hep G2 cells,the content of P-gp protein in He La cells was detected by immunofluorescence assay.6.Western blot was used to detect the content of MMP-2 protein in Hep G2 cells cells.7.Transwell assay was used to detect the migration ability of Hep G2 cells and He La cells.Results:1.The precipitate extracted by the kit exhibited a distinct lipid bilayer structure with a particle size of about 100 nm and expressed exosome marker proteins CD63,HSP70,and TSG101.2.Hematological malignancy serum-derived exosomes can carry P-gp protein into Hep G2 cells and He La cells,increase the content of P-gp protein in Hep G2 cells and Hela cells.3.Hematological malignancy serum-derived exosomes could induce Hep G2 cells and He La cells to be acquired resistance.4.Compared with the healthy human serum-derived exosomes,hematological malignancy serum-derived exosomes increased the content of P-gp protein in Hep G2 cells.5.Compared with the healthy human serum-derived exosomes,the content of P-gp protein was not significantly increased in exosomes derived from serum of hematological malignancy patients with chemotherapy.6.Hematological malignancy serum-derived exosomes could increase the migration ability of Hep G2 cells and He La cells.Conclusion:1.Hematological malignancy serum-derived exosomes could induce exogenous acquired resistance in Hep G2 cells and He La cells.2.Hematological malignancy serum-derived exosomes could increase the migration ability of Hep G2 cells and He La cells.
