Objective:It has been reported that immune cells play an important role in maternal and fetal tolerance and immune maintenance.However,the changes and role of immune cell subsets in preeclampsia have not been fully elucidated.The purpose of this study is to preliminarily explore the composition of immune cell subsets in preeclampsia and their changes and roles in the occurrence of preeclampsia(PE).Methods:Peripheral blood and decidua from severe PE patients(n=10)and normal pregnant women(n=19)were collected,Peripheral blood mononuclear cells(PBMCs)and decidual mononuclear cells were isolated.The ratio of CD16~+granulocytes(Gra),CD14~+mononuclear cells(Mon),CD163~+macrophages(Mac),CD56~+natural killer(NK)cells,peripheral blood and decidua was detected by flow cytometry.The expression level of CD158b,CD49a and CD103 in p NK,The ratio of CD49a~+CD39~+CD103~-d NK1 cells,CD49a~+CD39~-CD103~-d NK2 cells and CD49a~+CD103~+d NK3 cells in d NK(decidual natural killer)cells and the expression level of CD158b in d NK.In T lymphocytes,CD45RO~-CD45RA~+naive T cells(T_N),CD45RO~+CD45RA~-memory T cells(T_M)in CD3~+T cells,CD4~+T cells and CD8~+T cells,the level of PD-1 and CD161 in T cells,the ratio of TCR??~+??T cells.The percentage of CD25~+CD127~-Treg cells,and the level of CD39 they express.CD45,CD56,CD14,CD3,CD19 and FOXP3 protein expression levelaround decidual arteries was detected by immunohistochemical.Results:Compared with normal pregnant women,there was no significant difference in the ratio of CD14~+mononuclear cells,CD56~+NK cells,CD3~+CD4~+T cells,CD3~+CD8~+T cells and CD19~+B cells in the peripheral blood of PE patients to CD45~+DAPI~-immune cells.Almost no CD103 or CD49a was expressed in p NK cells,and there was no significant difference in the level of CD158b.The subpopulation ratio of T_N and T_M cells in the CD3~+,CD4~+,CD8~+T lymphocytes was not significantly different between the two groups;No significant difference was found in PD-1 or CD161 percentage of T cells;??T cell percentage,ratio of CD4~+CD25~+CD127~-Treg in CD4~+T cells and expression of CD39 had no differences in two groups.There were no significant difference in the large subsets of immune cells in the decidua of pregnant women with PE,also T lymphocytes,macrophages and NK cells were the main cells.And the proportion of CD49a~+CD103~+d NK3 cells in the d NK subgroup decreased significantly(P<0.05),but no significant difference was found in CD158b level.The subpopulation ratio of T_N and T_M cells in the CD3~+,CD4~+,CD8~+T lymphocytes was not significantly different between the two groups,the level of PD-1 and CD161 increased remarkably(P<0.05),There was a decrease in??T cells at the decidual junction(P<0.05),The proportion of CD4~+CD25~+CD127~-Treg cells in the deciduawas decreased(P<0.05)and CD39 expression decreased(P<0.05).The expression of CD56 and CD14 around decidual arteries in PE patients increased(P<0.05).Conclusion:Decreased proportions of CD49a~+CD103~+d NK3,??T cells and Treg cells in the decidua of PE patients may lead to the imbalance of immune toleranceinmaternal-fetal interface.Increased expression of PD-1 T lymphocytes in the decidua may lead to enhanced T cell killing function,Increased expression of CD161 in CD4~+and CD8~+T cells may lead to pro-inflammatory changes in the maternal-fetal interface,and decreased expression of CD39in decidual Treg may result in decreased Treg immunosuppression.These changes may have certain effects in the developmen of PE.
