| ObjectiveDue to the weakness of contraceptive awareness or improper contraceptive methods,many women use induced abortion as a remedy for contraceptive failure.However,many times of dilatation and curettage or hysteroscopy will lead to complications such as endometritis,intrauterine adhesions,and even lead to endometrial regeneration obstacles and difficulty of next pregnancy.Thin endometrium means that the thickness of endometrium is lower than the threshold thickness of embryo implantation.It has been pointed out that the thickness of endometrium is more than 7 mm,which is an important condition for embryo implantation.Many different therapeutic strategies have been explored,including vasoactive drugs,such as low-dose aspirin,sildenafil citrate,pentoxifylline combined with vitamin E,estrogen,granulocyte colony stimulating factor,growth hormone and regenerative medicine.However,these treatment methods have not formed the standard of treatment and scientific efficacy evaluation system.From the point of view of cost performance of pharmacoeconomics,vitamin E has a wide range of sources and low price.Many studies have shown that vitamin E alone or in combination has a good effect in the treatment of endometrial dysplasia,but it has not attracted enough attention in the clinical practice of thin endometrial treatment.In this experiment,we used the method of endometrial full-thickness resection to construct the model of thin type of endometrium in rats,and intervened with different doses of vitamin E to compare the expression of estrogen and progesterone,vascular endothelial growth factor(VEGF),CD31,interleukin-10(IL-10),leukemia inhibitory factor(LIF)in endometrium of rats in each group,and compared the thickness of endometrium and the number of glands in each group.It can provide animal experimental basis for clinical treatment.Methods:Y.Y.Kim etal established the model of endometrial injury in rats by mechanical injury,in this experiment,the thin endometrial model was established by the method of endometrial full-thickness resection.50 mature female Sprague Dawley rats of SPF grade were divided into normal group,model group,low dose group,middle dose group and high dose group by random digital table.The low dose group,middle dose group and high dose group were given vitamin E corn oil solution 15 mg/kg.d,30 mg/kg.d and 60 mg/kg.d respectively after the model operation,and the normal group and model group were given corn oil of the same volume.The cervical dislocation method was used to kill the rats in the estrous period after four estrous cycles.The uterine samples of each group were collected,the tissue sections and HE staining were used to observe the thickness of the endometrium and the growth of the glands.The expression of CD31 in the endometrium was detected by immunofluorescence method.The estrogen and progesterone,VEGF,CD31,IL-10 and LIF were detected by enzyme-linked immunosorbent assay.Results:Compared with the normal group,the endometrial thickness and the number of glands in the model group were significantly decreased(P<0.01),the expression of VEGF,LIF and CD31 in the endometrium was significantly decreased(P<0.01),and the endometrial thickness and the number of glands in the high dose group were significantly increased(P<0.01),and the expression of VEGF and CD31 were significantly increased(P<0.01).Compared with the model group,the endometrial thickness and the number of glands in the middle and high dose groups were increased significantly(P<0.01),the expression of VEGF,IL-10,LIF and CD31 in the middle and high dose groups was increased significantly(P<0.01),and the expression of estradiol and progesterone was increased with the increase of vitamin E dose(P<0.01)Conclusions1.The thin endometrium animal model was successfully made by endometrial full-thickness resection.2.Vitamin E may improve the local blood perfusion of endometrium,promote the growth and thickening of endometrium,and improve the receptivity of endometrium by up regulating the expression of CD31 and VEGF in endometrium and the secretion of estrogen and progesterone.3.Vitamin E can up regulate the expression of IL-10 to inhibit the inflammatory response of trauma,and improve the level of LIF,which is related to endometrial receptivity,so as to improve endometrial receptivity and play a repair role. |
