Objective:Detectioning expression of CK5/6,CD34 and Ki67 in breast cancer and breast fibroadenoma tissues by immunohistochemistry.Researching the relationship between the expression levels of CK5/6,CD34,Ki67 and clinicopathological factors in breast cancer tissues,and the correlation between the expression of CK5/6,CD34 and Ki67 in breast cancer tissue.Exploring the clinical significance of CK5/6,CD34 and Ki67 in treatment and prognosis of breast cancer.Method:Froming October 2018 to October 2019,150 patients who underwent modified radical mastectomy in our hospital were collected as the experimental group.Inclusion criteria: All of them were female without radiotherapy;chemotherapy;endocrinotherapy or targeted therapy;they were not double breast cancer and had no other tumor history.During the same period,20 patients with breast fibroma were used as the control group.The expression of CK5/6,CD34 and Ki67 in the experimental group and the control group was detected by immunohistochemistry.SPSS24.0 software was used to process the data.The ?2 test was used to compare the differences of the expression levels of CK5/6,CD34 and Ki67 in the two groups,and to analyze the relationship in the expression of CK5/6,CD34 and Ki67 in the experimental group with tumor size,histological grade,and lymph node metastasis or not and TNM staging.Spearman rank correlation analysis was used to explore the correlation of CK5/6,CD34 and Ki67 expression in the experimental group.Result:1 Expression of CK5/6,CD34 and Ki67 in experimental group and control group1.1 The positive expression rate of CK5/6 was 65.3% in 150 cases of breast cancer patients.The positive expression rate of CK5/6 was 20.0% in 20 cases of breast fibroadenoma patients.The difference of CK5/6 positive expression between the two groups was statistically significant(P < 0.001).1.2 The positive expression rate of CD34 was 76.7% in 150 cases of breast cancer patients.The positive expression rate of CD34 was 15.0% in 20 cases of breast fibroadenoma patients.The difference of CD34 positive expression between the two groups was statistically significant(P < 0.001).1.3 The highly expression rate of Ki67 was 58.0% in 150 cases of breast cancer patients.The highly expression rate of Ki67 was 15.0% in 20 cases of breast fibroadenoma patients.The difference of Ki67 high expression between the two groups was statistically significant(P<0.001).2 Relationship between expression of CK5/6,CD34,Ki67 and clini-copathological factors in experimental group2.1 The difference of CK5/6 positive expression among patients with or without lymph node metastasis groups was statistically significantly(P<0.001);The difference of CK5/6 positive expression among patients with different tumor sizes groups was no statistically significantly(P=0.812);The difference of CK5/6 positive expression among patients with histological grade groups was no statistically significantly(P=0.699);The difference of CK5/6 positive expression among patients with TNM staging groups was no statistically significantly(P=0.089).2.2 The difference of CD34 positive expression among patients with different tumor sizes groups was statistically significantly(P=0.007);The difference of CD34 positive expression among patients with histological grade groups was statistically significantly(P=0.016);The difference of CD34 positive expression among patients with or without lymph node metastasis groups was statistically significantly(P=0.002);The difference of CD34 positive expression among patients with TNM staging groups was statistically significantly(P=0.002).2.3 The difference of Ki67 high expression among patients with histological grade groups was statistically significantly(P<0.001);The difference of Ki67 high expression among patients with or without lymph node metastasis groups was statistically significantly(P=0.007);The difference of Ki67 high expression among patients with different tumor sizes groups was no statistically significantly(P=0.151);The difference of Ki67 high expression among patients with TNM staging groups was no statistically significantly(P=0.483).3 Correlation analysis of CK5/6,CD34 and Ki67 expression in experimental groupThe correlation between CK5/6 and CD34 expression in breast cancer tissues was no statistically significant(r=0.083,P> 0.05);The correlation between CK5/6 and Ki67 expression in breast cancer tissues was statistically significant(r=0.217,P<0.05);The correlation between CD34 and Ki67 expression in breast cancer was statistically significant(r=0.320,P<0.05).Conclusion:1 The expression of CK5/6,CD34 and Ki67 in breast cancer was signific-antly higher than that in fibroma,which proved that CK5/6,CD34 and Ki67 were significantly correlated with the incidence of breast cancer.2 CK5/6 is significantly correlated with lymph node metastasis of breast cancer;CD34 was significantly correlated with tumor size,histological grade,lymph node metastasis and TNM stage;The high expression of Ki67 was significantly correlated with breast cancer tissue grade and lymph node metastasis.CK5/6,CD34 and Ki67 may be involved in the evolution process of breast cancer,which may be helpful to predict the recurrence of breast cancer patients.3 There was not correlated between the expression of CK5/6 and CD34 in breast cancer;There was a positive correlation between the expression of CD34 and Ki67 in breast cancer,suggesting that angiogenesis and cancer cell proliferation were closely related to the invasion and metastasis of breast cancer;There was a positive correlation between the expression of CK5/6 and Ki67 in breast cancer,The combination of CK5/6 and Ki67 can improve the sensitivity and specificity of breast cancer prognosis evaluation and risk classification.Therefore,the combined detection of CK5/6,CD34 and Ki67 may be helpful for the clinical diagnosis and treatment of breast cancer patients and the evaluation of prognosis.
