Objective:To observe the effect of total glucosides of paeony on synovial injury in CIA rats,and to explore whether the effect of relieving RA inflammation may be realized by mucosal immunity by detecting the expression of IL-4,IFN-? and TGF-? in small intestinal mucosa and mesenteric lymph nodes of rats.Methods:1.8 of 72 clean SD rats were randomly selected as blank control group,and the rest were used to establish CIA model.The emulsion of bovine type ? collagen and complete Freund's adjuvant was injected into the caudal root and right hindfoot of rats at multiple points,and the immunity was enhanced 7 days later.After 7 days of enhanced immunization,the model rats were scored by AI,and the rat models with scores ? 6 were regarded as successful.Finally,40 of them were selected,and the success rate of modeling was 62.5%.They were randomly divided into 5 groups: model group,high,medium and low dose total glucosides of paeony treatment group and positive control group(sinomenine treatment group),with 8 rats in each group.The experimental dosage of total glucosides of paeony and sinomenine was converted according to the conversion formula of body surface area of experimental animals(the blank control group and model group were fed with the same volume of normal saline).All experimental animals were given medicine and diet regularly every day.The diet,hair luster,action state and mental state of the rats were observed,and the AI and the width of the right foot and ankle were measured every 4 days.After 28 days,the rats were euthanized,and the appropriate parts were selected for follow-up test.2.HE staining was used to observe the proliferation of cells and blood vessels in synovial tissue and the infiltration of lymphocytes in synovial layer and subsynovial layer.3.The expressions of IL-4,IFN-? and TGF-? in intestinal mucosa and mesenteric lymph nodes of rats were detected by immunohistochemistry.Summary:1.Total glucosides of paeony can significantly improve the arthritis of CIA rats,relieve the general inflammatory condition,and significantly reduce the arthritis index and ankle width of CIA rats.The therapeutic effect of high dose of total glucosides of paeony is the best,and it has better anti-inflammatory effect than the middle and low dose groups.2.Total glucosides of paeony can inhibit synovial inflammation to some extent,reduce lymphocyte infiltration and inflammatory substances in synovial tissue,and improve its proliferation.3.Total glucosides of paeony can promote the expression of IL-4 and TGF-? in intestinal mucosa and mesenteric lymph nodes of CIA rats,and inhibit the expression of IFN-?.The promotion or inhibition effect of high dose TGP group is the most obvious,but there is no sufficient evidence to prove that this effect is related to the dose of total glucosides of paeony.4.The inhibitory effect of total glucosides of paeony on joint inflammation in CIA rats may be related to its regulatory effect on the expression of IL-4,IFN-? and TGF-? in small intestinal mucosa and mesenteric lymph nodes.
