| Background and objective: In this experiment,collagen-induced arthritis(CIA)rats were used as models,and total glucosides of paeonia(TGP)was intragastric dosing to observe the effects of TGP on feces of CIA rats,and on serum 5-HT,VIP,colon AQP3,5-HT1 A receptors.The effect of 5-HT,5-HT1 receptor,to explore the mechanism of TGP induced changes in feces traits of rats.Methods: 70 male SD rats were randomly selected as control group(8 healthy rats)and other rats as model group.The unsuccessful rats were eliminated,and 40 rats with successful models were randomly divided into 5 groups with 8 rats in each group.They were model group,low-dose TGP group,middle-dose TGP group,high-dose TGP group and sinomenine group.Each group was administered by intragastric dosing.Control group and model group were given normal saline,and TGP were prepared with normal saline to the required concentration for intragastric dosing.Sinomenine group was given sinomenine.During the experiment,the general condition,weight,fecal water content,weight of fecal,arthritis index,and scores of arthritic flexion pain test of the rats were observed.The samples were taken for experiments on the next day of the final administration.The serum 5-HT and VIP levels,colon AQP3 and 5-HT1 A receptor levels were measured,and colon pathological staining was performed.The effects of TGP on the above indexes were statistically analyzed.Results: 1.TGP can improve the arthritis of CIA rats,reduce arthritis index,the width of the ankle,and the scores of arthritic flexion pain test of CIA rats;2.High and middle dose TGP can cause diarrhea and increase fecal water content in rats;3.TGP can increase the concentration of 5-HT and VIP in serum of CIA rats,up-regulate5-HT1 receptor and down-regulate AQP3 expression in colon of CIA rats.Conclusion: A certain concentration of TGP can cause diarrhea and fecal shape changes while improving the arthritis of CIA rats.Its mechanism may be related to influence the gastrointestinal hormone. |
