Objective:The research is to study the function of the hypothalamus during the central response of acupuncture regulating the stomach function,through analyzing the change of significant metabolite in the hypothalamic(HYP)zone of the Functional Dyspepsia model rat with H Proton Magnetic Resonance Spectroscopy(1H-MRS).Methods:1.30 healthy Spargue-dawley male rats(7-days-old)were divided into three groups: control group,model group and acupuncture group with random method.2.Process the rats of model group and acupuncture group with 0.1%iodoacetamide gavage,tail clamping and irregular feeding to induce FD model.The general condition,the change of weight,foot intake and water intake were recorded at the same time.3.The rats of acupuncture group were electroacupunctured for ten days under binding after the model making stage,while the rats of control and model were bound in the same with no electroacupuncturing.4.After the intervention stage,all the rats were treated by starch gavage and the stomach were taken out after a while to detect the gastric emptying rate.Meanwhile,the blood of precava were drawn to test the gastrin(GAS)in serum with ELIAS analysis method.5.The brain of the rats were taken out quickly to perform the 1H-MRS examination and get the absolute concentrations of N-acetylaspartate(NAA),Aspartic Acid(Asp),Glycine(Gly),etc.through the area under the peak calculated by integration method,with the help of TOPSPIN software.Results:1.After the modeling stage,the general condition,weight,food intake and water intake of the model group rats and acupuncture rats were significantly lower than the control group rats(P<0.05).2.After the intervention stage,weight changes,food intake,water intake and gastric emptying rat significantly increased and GAS level decreased compared with the model group rats(P<0.05).3.After the intervention stage,the Myo concentration in hypothalamus of the acupuncture group rats increased,and the difference was statistically significant compared with the model group(P<0.05).The Myo and Ala concentration in hypothalamus of the model group rats decreased,and the difference was statistically significant compared with the control group(P<0.05).Conclusion:1.Acupuncture on the stomach Shu-Mu points could obviously improve the general condition,weight,foot intake and water intake and regulate gastric emptying rate and gastrin level at the same time.2..Acupuncture on the stomach Shu-Mu points could obviously change the Myo concentration in the hypothalamus of the FD rats,which may be one of the mechanisms of acupuncture treating the FD through regulating the function of the hypothalamus.
