Objectives Toll-like gene,a kind of protein molecule,plays an very important role in non-specific immunity,and also serves as a link between specific and non-specific immunity.The TLR4 gene is an important part of the Toll-like gene family.TLR4 has a significant relationship with the occurrence and development of many tumors,this study investigates the relationship between single nucleotide polymorphism of TLR4 gene rs7873784 and susceptibility to liver cancer.Methods A case-control study was used.The study subjects included 240 liver cancer patients and 500 normal case as control.The patient group was diagnosed by pathological examination from 2011 to 2015,all the patients come from Tangshan Workers Hospital Affiliated to North China University of Technology and Tangshan People 's Hospital Affiliated to North China University of Technology.None of them had received radiochemotherapy or targeted therapy,they had no history of other malignancies.The control group randomly selected 500 healthy people in Tangshan area who had no blood relationship with the case group during the same time.Taqman probe method was used to analyze the genetic variation of 3?UTR-rs7873784 of the TLR4 gene,and the odds ratio(OR)and 95% confidence interval(CI)were calculated by unconditional logistic regression to evaluate the different genotype carriers the relative risk of liver cancer.Results TLR4 rs7873784 GG and GC + CC genotypes accounted for 92.5%(222/240)and 7.5%(18/240)in the case group,and TLR4 rs7873784 GG and GC + CC genotypes accounted for 84.8%(424/500)and 15.2%(76/500)in the control group,after unconditional logistic regression analysis,compared with rs7873784 GG genotype carrier,GC + CC genotype reduces the risk of liver cancer(OR = 0.450,95% CI = 0.262-0.773,P = 0.004).Taking the GG genotype as a reference,in the gender stratification,the GC + CC genotype reduced the risk of liver cancer in male patients by OR(95% CI)0.411(0.223-0.756)(P <0.05).Age stratification showed that GC + CC genotypes had a lower risk of liver cancer than carriers of GG genotypes in the> 60-year-old group(OR = 0.339,95% CI = 0.136-0.849,P = 0.021).In the smoking stratification,compared with the GG genotype,the GC + CC genotype reduced the risk of liver cancer in the non-smoker group(OR = 0.270,95% CI = 0.125-0.582,P = 0.001).In the drinking stratification,compared with the GG genotype,the GC + CC genotype reduced the risk of liver cancer in the non-drinker group(OR=0.342,95%CI=0.185-0.633,P=0.001),In the presence or absence of hepatitis B stratification,compared with GG genotype,GC+CC genotype reduces the risk of hepatitis group(OR=0.45,95%CI=0.193-0.768,P=0.024).Conclusions The genetic variation of TLR4 rs7873784G> C reduces the susceptibility of liver cancer.Figure1;Table3;Reference 191...
