| Background Podocyte injury is the key factor affecting the progression of diabetic nephrology(DN),however there is no effective way to prevent or cure it yet.Objectives To explore the curative effect and possible mechanisms of AG plus NG on podocyte injury in DN,and provide new ideas for the clinical prevention and treatment of DN.Methods In this study,diabetic rats were treated with AG,NG,and AG plus NG respectively for 12 weeks.Albuminuria,renal histopathology and podocyte ultrastructure were measured,as well as podocyte related proteins,oxidative stress,podocyte mitochondria injury and podocyte apoptosis.To uncover the underlying mechanisms of AG plus NG on DN rats,the activity of LKB1/AMPK/Fox O1 pathway was detected.Results AG plus NG synergistically reduced albuminuria,improved renal pathological,ameliorated podocyte apoptosis and detachment,ameliorated mitochondrial injury and oxidative stress,as well as regulated the activation of LKB1/AMPK/Fox O1 pathway.Conclusion AG plus NG synergistically ameliorated podocyte apoptosis and detachment,the effects of which were related to the alleviation of oxidative stress and mitochondrial dynamic dysfunction,as well as regulation of LKB1/AMPK/Fox O1 pathway. |
