Objectives1.The serum adiponectin(APN)levels were detected between patients with missed abortion and normal pregnant women to study the relationships between serum APN level and missed abortion in early pregnancy.It may provide a new direction for the understanding and prediction of missed abortion.2.The expressions of adiponectin receptor 1(Adipo R1)and adiponectin receptor 2(Adipo R2)were detected in the chorionic villi between patients with missed abortion and normal pregnant women to explore the effects of the expression of adiponectin receptor(Adipo R)on missed abortion in early pregnancy.It may improve the understanding of the pathogenesis of missed abortion and provide more rich reference materials.Materials and methods1.Objects of study The serum and chorionic tissues were collected,including 28 patients with missed abortion in early pregnancy and 30 women of early normal pregnancy who decided voluntarily to interrupt their pregnancy.2.Experimental grouping The serums and villis of 28 patients with missed abortion were enrolled as missed abortion group.The serums and villis of 30 early normal pregnancy women were enrolled as normal pregnancy group.3.Experimental methods Immunohistochemistry was used to detect the expressions of Adipo R1 and Adipo R2 in the villis between the missed abortion group and the normal pregnancy group.The image integral optical density(IOD)was measured by the image analysis system(Image-pro plus 6.0)to indicate the expression intensity of each index.ELISA method was used to detect the serum APN concentration between patients with missed abortion and the early normal pregnant women.4.Statistical analysis The experimental data were expressed as mean ± standard deviation(SD).The statistical data was analysed with Independent-samples t test and Person correlation analysis by SPSS 13.0statistical software.A P value < 0.05 was considered to be significant.Results1.Serum APN levels between patients with missed abortion and normal pregnant women during early pregnancy The average serum APN concentration in missed abortion group(44.42 ±25.36 ug / ml)was lower than that in normal pregnancy group(57.83±29.79 ug / ml),but there was no significant difference in statistics(P > 0.05),which might indicat that the serum APN level in early pregnancy had no effect on the occurrence of missed abortion.2.The expressions of Adipo R1 and Adipo R2 in chorionic villi between patients with missed abortion and normal pregnant women during early pregnancy Adipo R1 mainly expressed in cytoplasm of trophoblastic layer in normal pregnancy group while expressed in nucleus of cytotrophoblast layer in missed abortion group.The expressions of Adipo R1 in cytotrophoblastic layer were higher than that in syncytiotrophoblastic layer.Adipo R2 in villi were mainly distributed in trophoblastic layer in two groups,and expressed both in nucleus and cytoplasm.The average IOD(19698.82 ±11606.42)of Adipo R1 in missed abortion group was significantly lower than that in normal pregnancy group(24069.22 ±11420.36).Meanwhile,the average IOD(15458.79 ±6335.97)of Adipo R2 in the missed abortion group was significantly lower than that in the normal pregnancy group(17792.92 ±6161.98).There was a significant difference both in Adipo R1 and Adipo R2 between missed abortion group and normal pregnancy group(P < 0.05).Conclusions1.The average serum APN concentration in missed abortion group was lower than that in normal pregnancy group,but there was no significant difference in statistics.This suggested that the serum APN level might can't be used as an index for helping to diagnose missed abortion in early pregnancy.2.Adipo R1 and Adipo R2 were expressed in placental villi of early pregnant women.The Adipo R1 and Adipo R2 of villi were mainly distributed in trophoblast layer and a little in villus stroma.3.The expressions of Adipo R1 and Adipo R2 in chorionic villi of patients with missed abortion was significantly lower than that in normal pregnant women during early pregnancy,which suggested that the lower expressions of Adipo R1 and Adipo R2 were one of the reasons for the formation of missed abortion in early pregnancy.
