| Objective: To investigate whether miR-455 is involved in the regulation of ERS-mediated apoptosis by hydrogen sulfide.Methods: The resuscitated epidermal cells were used as research subjects and cultured into the third generation,grown in 96-well plates for 24 hours.Cultured epidermal cells were randomly divided into three groups: group of miR-455 inhibitor,group of miR-455 inhibitor and group of negative control.The epidermal cells of three groups were transfected with miR-455 mimic,miR-455 inhibitor and miR-455 negative control.After 24 hours,the three groups were cultured in a medium of 10 % burn serum.The process takes four hours.The epidermal cells from the above process were cultured in a 10%fetal bovine serum with 150mol/L NaSH for 16 hours.qPCR was used to detect the relative mRNA expression levels of Caspase-12 and CRT,Western blot assay was used to detect the protein expression level of Caspase-12 and CRT.Results: 1.The expression of CRT mRNA was decreased in the three groups,and there was significant difference in them respectively(p<0.05).2.mRNA and protein expression levels of Caspase-12,CRT were significantly different in each group(p<0.05).All three groups were treated with Hydrogen Sulfide after burned rat serum on the epidermal cells damage.Conclusions: 1.With the intervention of exogenous hydrogen sulfide,the expression level of mRNA of Caspase-12 in epidermal cells which were damaged by burned serum is directly proportional to the expression level of miR-455,this result indirectly reflects that the decreased expression of miR-455 can reduce endoplasmic reticulum stress in rat epidermal cells after burns.2.The expression of miR-455 plays an important role in the protection of rat epidermal cells with exogenous hydrogen sulfide after burn. |
