| Objective: The liquid chromatography-mass spectrometry(LC-MS)was used to detect the blood samples from patients with early colorectal cancer,advanced colorectal cancer and healthy people,to distinguish the differences in their metabolites,and to find the diagnosis of early colorectal cancer.Provide scientific basis for important potential biomarkers.Method: The blood samples of 16 patients with early colorectal cancer,22 patients with advanced colorectal cancer and 20 healthy people were detected by liquid chromatography-mass spectrometry(LC-MS).Compare the results of total ion current diagram and principal component analysis of three groups of samples,and observe whether there are differences among the three groups of samples.The total ion flow chart and principal component analysis results of the three groups of samples were compared,and through variance analysis,partial least squares discriminant analysis,ROC curve,cluster analysis and other methods,differential metabolites with strong discrimination ability were screened out.After that,the mass nuclear ratio of the metabolites was introduced into the human metabonomics database(HMDB)to identify the structure and explain its biological significance.Result: 1.Twelve kinds of differential metabolites were found in the analysis of blood metabolism map of early stage colorectal cancer patients,advanced stage colorectal cancer patients and healthy people,and these metabolites were able to distinguish well between the three groups.2.Six different metabolites,Melatonin B,lysophosphatidylcholine,lysophosphatidic acid,lysophosphatidylethanolamine,androsterone sulfate and arginine,were found in the blood metabolism map analysis of colorectal cancer patients and healthy people.The expression of Melatonin B,lysophosphatidylcholine,lysophosphatidylethanolamine and lysophosphatidic acid showed an increasing trend in the colorectal cancer group The expression of androsterone sulfate and arginine decreased in colorectal cancer group,suggesting that these six metabolites may be potential biomarkers for colorectal cancer.3.Three metabolites,melatonin B,lysophosphatidylcholine and lysophosphatidylic acid,were found to be significantly different in the blood metabolic map analysis of patients with early colorectal cancer and healthy people.The expression of melatonin B,lysophosphatidic acid was on the rise,and lysophosphatidylcholine was on the rise first and then on the decline,suggesting that these three metabolites may be potential biomarkers for early colorectal cancer.Conclusion: 1.There are different metabolites in the serum of early colorectal cancer patients and advanced colorectal cancer patients and healthy people;2.The six metabolites of melatonin B,lysophosphatidylcholine,lysophosphatidic acid,lysophosphatidylethanolamine,androsterone sulfate and arginine showed significant differences between the colorectal cancer group and the healthy group.3.The three metabolites of melatonin B,lysophosphatidylcholine and lysophosphatidic acid showed significant differences between the early colorectal cancer group and the healthy group,which may be potential biomarkers for early colorectal cancer diagnosis. |
