Objective: To investigate the correlation between follicular fluid(FF)high density lipoprotein(HDL)granule composition and embryo quality in vitro fertilization(IVF)patients.Methods: From August 2019 to March 2020,504 follicular fluid samples from the infertile women who received IVF-ET in our department were enrolled in the study,who met the inclusion criteria but did not exclude the criteria.The levels of apolipoprotein A-1(ApoA1)and apolipoprotein B(ApoB)in follicular fluid were measured by immunoturbidimetry,and the levels of HDL subclasses in follicular fluid were determined by immunoblotting.The levels of FSH,LH,HCG and anti-Mullerian hormone in follicular fluid were detected by epidemiological analysis;the contents of ROS in follicular fluid were detected by human reactive oxygen species ELISA kit,and the total antioxidant capacity Kit was used.Total antioxidant capacity(TAC)of follicular fluid was measured.Transplantable embryo were divided into high-quality embryos and non-high-quality embryos according to whether they were high-quality embryos.The levels of ApoA1,HDL subclasses in follicular fluid of the two groups were observed.Regarding the quality of embryos as strain variable,the independent variable was introduced into the binary multivariate Logistic analysis to observe the correlation between HDL components and embryo quality.Results: A total of 266 high-quality embryos were obtained,and the rate of high-quality embryos was 25.88%.There was no significant difference in FSH level between superior and non-superior follicular fluid(P > 0.05).LH level in superior follicular fluid was significantly lower than that in non-superior follicular fluid;HCG and AMH levels were significantly higher than that in non-superior follicular fluid(P < 0.05).The level of ROS in follicular fluid of the eugenic group was lower than that of the non-eugenic group(P < 0.05),and the level of TAC in follicular fluid of the eugenic group was higher than that of the non-eugenic group(P < 0.05).The level of ApoA1 in follicular fluid of eugenic group was higher than that of non-eugenic group(P < 0.05),and the levels of ApoB in follicular fluid of eugenic group werelower than those of non-eugenic group(P < 0.05).The levels of prebeta 1-HDL,HDL3 c and HDL3 b in follicular fluid of high-quality embryo group were lower than those of non-high-quality embryo group(P < 0.05),and the levels of prebeta2-HDL in follicular fluid of high-quality embryo group were lower than those of non-high-quality embryo group,but there was no significant difference(P >0.05).The levels of HDL3 a,HDL2a and HDL2 b in the follicular fluid of the superior embryo group were higher than those of the non-superior embryo group(P < 0.05).The spearman correlation analysis showed that there was no correlation between FSH,prebeta 2-HDL levels and eugenic embryos(P > 0.05),and the levels of LH,ROS,ApoB,prebeta 1-HDL,HDL3 c,HDL3b in follicular fluid were negatively correlated with eugenic embryos(P<0.05),while the levels of HCG,AMH,TAC,ApoA1,HDL3 a,HDL2a,HDL2 b in follicular fluid were positively correlated with euembryos(P<0.05).The multivariate logistic analysis showed that ApoA1,HDL2 b and preβ1-HDL were the influencing factors for eugenic embryos(P<0.05).Conclusion: The correlation between follicular fluid(FF)high density lipoprotein(HDL)granule composition and embryo quality were confirmed.ApoA1,HDL2 b and preβ1-HDL in HDL granules have predictive for embryo quality,which is worthy of further clinical study.
